| Background:The patients of chronic eczema make up a significant portion of the dermatological patients. The disease caused by many complicated factors includes internal and external factors, which dose not respond to standard treatment. In clinical, chronic eczema marked by frequent relapse and unable to resistant itch. The main treatment of localized chronic eczema is topical preparation. Western medicine commonly uses steroids, which have many side effects. For the good of severing the patients, we distribute to research. Therefore, we need to seek more effective topical products to meet the clinical use. It is the most important and first work in research. Densefruit Pittany Root-bark show high possibility in allergic diseases. In present, Densefruit Pittany Root-bark is only to be used in compound agents in the treatment of chronic eczema. It requires further research to test its effective components and the mechanism for curing eczema.Objective:This research is for the purpose of confirming the effect from treating the chronic eczema with the extract of Densefruit Pittany Root-bark, and to explore its action mechanism. In the meanwhile, offering experimental data to the research of its effective components.Study process and method:1 Effect of Densefruit Pittany Root-bark on antipruritic (histamine to send itches response)Take the white guinea pigs, then randomly divided into positive control group, model group, normal dose group,2times dose group and 4times group of Densefruit Pittany Root-bark ethanol extract(n=10 each).At first, abraded the right dorsum of foot of the guinea pigs, 10min latter, started dropping 0.01% histamine in the injured area, 0.05ml each guinea pig, Hereafter increased progressively gradually the density, each time 0.05ml each guinea pig, until the guinea pig returned to lick the right foot. Then use the total dosage of Histamine as the final itching threshold. Then compare the total dosage of each group. By means of painting extract of Densefruit Pittany Root-bark, we observe the itching reaction of Guinea pig and calculate the itching threshold. Take all of this above; we can see the effect of extract of Dense fruit Pittany Root-bark for cure itch.2 Effect of Densefruit Pittany Root-bark on Guinea pigs eczema model induced by DNCB.Select the white guinea pigs, then randomly divided into positive control group, model group, normal dose group, 2times dose group and 4times group of extract of Densefruit Pittany Root-bark (n=10 each).First, use DNCB as the offending substance to induce allergic contact dermatitis on Guinea pigs. After making the DTH model, guinea pigs were applied the medicine, twice one day, continual thirty days. Finally execute the guinea Pigs, cut down their right ears, then observe the histopathological change and the inflammatory cell infiltration. The DTH Guinea pig model established by DNCB is used to test the effect of Densefruit Pittany Root-bark.3The 5-HT in affected part of DTH Guinea pig model established by DNCB is tested by immunohistochemical method.The study is used to test 5-HT in the affected part of Guinea pig with chronic eczema by immunohistochemical method. The aim of the research is test the effect of Densefruit Pittany Root-bark to 5-HT.Results:The monomers extract of Densefruit Pittany Root-bark (Fraxin-ellone,dictamnine,Obacunone)could obviously increase the itch threshold induced by histamine in guinea pigs. The monomers extract of Densefruit Pittany Root-bark extracthad a significant suppressive effect on DTH in Guinea pig, and it can relieve inflammatory cellular induced by DNCB. The monomers extract of Densefruit Pittany Root-bark can also inhibit the release of 5-HT which is a mediators of inflammation act in the process of DTH.Conclusion:The monomers extract of Densefruit Pittany Root-bark has notable function-on reliving inflammation, hypersensitivity and itching. It is also can inhibit the release of 5-HT. Overall, the monomers extract of Densefruit Pittany Root-bark has certain effect on the treatment of chronic eczema. |
