Experimental Study On Anti-animal Model Of Colorectal Cancer Efficacy Of Extracts Of Prescription Of TCM AK Combined With Cisplatin

[Objective]:To investigate the synergistic effects of cisplatin (CDDP) combined with the extracts of AK prescription on the proliferation of CT-26 cancer of colon cells, and to investigate the toxicity-reducing and synergistic effects of the extracts of AK prescription for chemotherapy.[Methods]:1. MTT (Methy thiazolyl tetrazolium) assay was used to evaluate the inhibitory effects of CDDP combined with the extracts of AK prescription on cells CT-26 in different time and different concentration.2. Models of Colorectal CT-26 tumor bearing mice were established and randomly divided into six groups, including group A (control group), group B (CDDP group), group C (AK group) and combined treatment groups D, E and F. The medication was begun after 24 hours since inoculation, once a day for 12 days. Mice were sacrificed on day 14th.The tumor inhibitory rate, thymus index, spleen index, liver and renal function and the blood cell in the peripheral blood were measured. The subtypes of T lymphocytes in the peripheral blood were detected by flow cytometry. Optical microscopy was employed to observe the pathomorphology of tumor and renal.[Results]: 1. CDDP combined with the extracts of AK prescription could inhibit the proliferation of the CT-26 cancerous cells in vitro.2. The tumor weight in groups D, E and F was lighter than that in group A (P<0.05). Groups B, C, D, E and F could inhibit the tumor growth and the tumor inhibition rate was 55.92%,28.11%,61.05%,71.11%,69.72%, respectively.3. Compared with group A, in group B the count of WBC and PLT was decreased obviously and the level of blood urea nitrogen (BUN) was increased (P<0.01). Compared with group B, in groups C, D, E and F the count of WBC and PLT was increased and the level of BUN was decreased (P<0.01).4. Compared with group A, in group B the level of CD3+,CD4+,CD4-/CD8+was lower and the level of CD8-was higher (P<0.05).The level of CD3+,CD4+,CD4+/CD8+ in groups C, D, E and F, was higher than that in group B, but CD8+ value was decreased (P<0.01).5. Compared with group A, thymus index and spleen index were decreased in group B (P<0.05).Groups C,D, E and F had different levels of increase in thymus index and spleen index compared with group B (P<0.05).6. Compared with group A, the mice in group B were got lesion in kidney pathological. Groups C, D, E and F had different levels of lessening in the injury of renal histology compared with group B.[Conclusion]:1. CDDP combined with the extracts of AK prescription could inhibit the proliferation of the CT-26 cancerous cells in vitro.2. The extracts of prescription of TCM AK can display the toxicity-reducing and synergistic effects for chemotherapy. Its mechanisms may be related to several functions such as anti-tumor, improving the immune function, protecting hematopoietic function and antioxidant.
