Objective To study the therapeutic effects of sodiumβ-aescin and panax notoginseng saponins(PNS) on cerebral edema,the permeability of blood-brain barrier(BBB) and metalo-proteinases-9 (MMP-9) in rats brains following intracerebral hemorrhage(ICH).Methods1.24 male adult SD rats were randomly divided into three groups, the rnegadose of sodiumβ-aescin and PNS group, the mediandose of sodiumβ-aescin and PNS group and the smalldose of sodiumβ-aescin and PNS group. The model of intracerebral hemorrhage was established by Rosenberg,s method, and rats were injected intraperitoneally corresponding medicine in different groups at every other 24h after operation. Then the rats were decapitated at 72h after intracerebral hemorrhage,and brains were taken for examining the brain water content.2.120 male adult SD rats were randomly divided into five groups, the sham-operated group,the model group,the PNS group,the sodiumβ-aescin group and the sodiumβ-aescin and PNS group. The five groups were further divided into the groups of examining the brain water content, examining the permeability of blood-brain barrier and examining the MMP-9. The model of intracerebral hemorrhage was established by Rosenberg,s method, and rats were injected intraperitoneally corresponding medicine in different groups at every other 24h after operation. In sham-operated group and the model group, equally dose saline was injected intraperitoneally.3. The neurofuction of rats were evaluated at 6h,12h,24h,48h,72h after intracerebral hemorrhage;4. The rats of examining the brain water content group were decapitated at 72h after intracerebral hemorrhage, and the brain water content was measured by using the dry-wet weight assay.5. EB solution,which cycles in vivo for 2 hour, was given through vena caudalis separately at 70 hours post-operation, and then the rats of examining the permeability of blood-brain barrier group were killed and brains were taken for the detection of the contents of EB in brain tissue.6. The rats of examining the MMP-9 group were decapitated at 72h after intracerebral hemorrhage, and rats brains were taken for histopathological and hematoma size examinations. Positive reacted cells of MMP-9 are counted under light microscope. Results1. At 72h after the operation, brain water content of the megadose of sodiumβ-aescin and PNS group was lower than that of other groups(p<0.05).2. Nurobehavioral scores were decreased in the rats with intracerebral hemorrhage; As compared to the sham-operated group, neurobehavioral scores were lower in the model group at every time points (p<0.05). However, neurobehavioral scores incereased gradually after the treatment of drug injection; Scores of drug groups were higher than that of the model group(p<0.05); And in the drug groups, scores of the sodiumβ-aescin and PNS group were higher than that of other groups(p<0.05).3.The examination of histopathological, water content,hematoma size, permeability of blood-brain barrier show that the intracerebral hemorrhage damage in the PNS group, the sodiumβ-aescin group,the sodiumβ-aescin and PNS group was significantly milder than that in the model group(p<0.05). Positive immune reacted cell amounts of MMP-9 were alleviate in the sodium P-aescin and PNS group as compared with the sodiumβ-aescin group and the PNS group at 72h(p<0.05).Conclusions1. The preparation of this ICH rat model, established by stereotaxic injection of collagenaseⅦ, was relative easy, reliable and practical.2. The cooperation protective effect of sodiumβ-aescin and PNS injection therapy against cerebral edema following intracerebral hemorrhage might be associated with alleviating the positive expressions of MMP-9. Moreover, the cooperation therapeutic effect of sodium p-aescin and PNS is superior than the unity therapeutic effect of sodiumβ-aescin or PNS.
