Objective To know if there are some differences of Behavior and the Glu,GABA concentration of cortex,hippocampus and the Treatment effects of valproate (VPA) between Penicillin (PEN) and Pentylenetetrazole (PTZ) induced acute seizure model rats. Methods 63 female rats were randomly assigned to five groups (n=10, rats which were dead before the predetermined time were excluded from the groups):the control group, PEN model group (PEN model), PTZ model group (PTZ model), PEN model VPA pretreated group (VPA+PEN group), and PTZ model VPA pretreated group(VPA+PTZ group). The Behavior of rats were recorded by camera. Immunohistochemical staining of SABC method and High Performance Liquid Chromatography-Mass Spectrometry-Mass Spectrometry (HPLC-MS-MS) method were chose to determine the concentration of Glu and GABA of cortex and hippocampus of these rats. Used statistical software spss13.0 to compare the datas of the latency of seizures,seizures score and the Glu and GABA concentration of cortex and hippocampus between the control group and the two model groups and between the two model groups and between the two model groups and their respective VPA pretreated groups.Results Rats from the control group behaved normal, PEN group behaved quiet; PTZ group behaved excited during the latency of seizures. Rats from PEN group got a more longer latency of seizures compared with PTZ group (p<0.05). Both of the Rats from the PEN and PTZ groups could appearedâ…¢-â…¤levels seizures (according to Racine score).VPA can prolonged the latency and lower the levels of seizures both of PEN and PTZ models (P<0.05). The Glu concentration of the cortex and hippocampus of rats from the PEN group was higher than that of the control group (P<0.05), GABA concentration was lower than the control group (P<0.05). The Glu concentration of the cortex of rats from the PTZ group was higher than that of the control group (P<0.05); concentration of the hippocampus had no difference with the control group (P>0.05). The GABA concentration of the cortex and hippocampus of rats from the PTZ group was higher than that of the control group (P<0.05). The Glu concentration of the cortex and hippocampus of rats from the VPA+PEN group was lower than that of the PEN group (P<0.05), GABA concentration was higher than the PEN group (P<0.05). The Glu concentration of the cortex of rats from the VPA+PTZ group was lower than that of the PTZ group (P0.05); concentration of the hippocampus had no difference with the PTZ group (P>0.05). Conclusions 1 Rats from PEN group got a more longer latency of seizures compared with PTZ model; rats from PEN model behaved quiet; PTZ model behaved excited during the latency of seizures.2 The Glu concentration of the cortex and hippocampus of rats from the PEN group was increased, GABA concentration was decreased. The Glu concentration of the cortex of rats from the PTZ group was increased, concentration of the hippocampus had no change. The GABA concentration of the cortex and hippocampus of rats from the PTZ group was increased.3 VPA can prolonged the latency and lower the levels of seizures and reduced the amplitude of variation of Glu and GABA concentration of the cortex and hippocampus both of the PEN and PTZ acute seizures models(except for Glu concentration of rats from PTZ model).4 PEN seizures model is similar to human absence seizures.PTZ seizures model is similar to human temporal lobe seizures,it is a perfect temporal lobe seizures for research.
