ObjectiveCurrently, Modern Medicine considers the cause of depression is monoamine hypothesis. Lot of antidepressant in use are mostly based on the pathogenesis of depression monoamine hypothesis, which often show some obvious flaws, like Slow onset, after drug withdrawal symptoms, drug dependence and so on. Many studies have shown that patients with depression often show a clear circadian rhythm disorder.Thus biological rhythm disorders may be one important component of depression. It is good for a formation new hypothesis. Therefore, we propose that made the circadian rhythm Normalization is new antidepressant strategy, Inorder to perspective the effect of liver-regulating compound recipe JWSNS Decoction in antidepressant.Methods1. The effect of liver-regulating compound recipe Jiawei Sins Decoction in rat circadian rhythmUse Willner P modeling with an unpredictable chronic mild stress model and treating the rats by JWSNS along with the copying.2. DetectionQuantitative PCR:at the end of the modeling period, all rats were died and took its baind, the time were 10:00,14:00,18:00,22:00,02:00 and 06:00. Take the SCN from rats blind and keep in the liquid nitrogen. Use Real-time PCR to test gene circadian rhythm's Save volume of SCN include Bmall, Clock, cry, per, and observe the changes of it.The Changes of behaviouristics:after replication model we use the Clock Lab running wheel system for circadian biology to fine its Rho, Onset (hr), Acrophase (hr), Total (hr). Finally, analysis the changes of behaviouristics and explore the generation mechanism. Results1 Changes in gene circadian rhythm by CMUS(1) Comparison of different time:Experimental results show that in the regulation of Bmall gene, Clock gene, Per1 gene, Per2 gene, Per3 gene, Cry2 gene caused by chronic stress, the 2ΔΔCT value is rise in the most time.(2) Fitting the data obtained diurnal variation of different time:CMUS could lead to an average of 2ΔΔCT value, peak and valley rise, fluctuations become larger, phase advance with Bmall gene and gene circadian rhythm.Because of an average of Perl gene's 2ΔΔCT value, peak and valley chaos Cannot compare fluctuations and phase. CMUS could lead to an average of 2ΔΔCT value and peak rise,fluctuations become larger,phase postponed, but not valley with Per2 gene. CMUS could lead to an average of 2ΔΔCT value drops, peak rise,fluctuations did not change, phase postponed, but not valley with Per3 gene. CMUS could lead to an average of 2ΔΔCT value, peak and valley rise.fluctuations did not change.phase postponed with Per3 gene. CMUS could lead to an average of 2ΔΔCT value and peak rise, fluctuations and phase did not change, but not valley with Cry2 gene.2 Changes in behaviouristics by CMUSThe results showed that Acrophase (hr) postponed, Rho and Total (hr) rised Onset (hr) advanced.3. Jiawei Sini Decoction and positive inference mechanism of antidepressant drugs melatonin in gene circadian rhythmMao Shi group and You Shi group mainly through changes in gene content and circadian rhythm to achieve antidepressant effects. They can show a regulation of gene circadian rhythm,average of 2ΔΔCT value, peak and valley rise, fluctuations。Due to Antidepressant drugs melatonin have bad effect in gene circadian rhythm, we reasoned that antidepressant drugs melatonin can't regulate the changes of gene circadian rhythm. Antidepressant drugs melatonin achieve antidepressant effects no regulate the changes of gene circadian rhythm, but another one. May be in regulation of time, which is the early experiments.Because of Mao You Shi time share group show a bad regulation of gene content and rhythm to achieve antidepressant effects.4. Jiawei Sini Decoction and positive inference mechanism of antidepressant drugs melatonin in behaviouristics. With the changes of Rho,Onset (hr), Acrophase (hr), Total (hr) by chronic stress, Liver Decoction Jiawei Sini Decoction and positive inference mechanism of antidepressant drugs melatonin show a good regulation. You Shi group is the most effect in those group.Conclusions1 CMUS can bring the disorder of gene circadian rhythm, including Bmall gene, Clock gene, Perl gene, Per2 gene, Per3 gene, Cryl gene, Cry2 gene.2. Mao Shi group and You Shi shows a good regulation of gene by chronic stress. But Mao You timeshare group and antidepressant drugs melatonin can't3.Decoction Jiawei Sini Decoction Mao Shi group and You Shi shows a good regulation of Circadian rhythms of gene include amplitude and phase.But Mao You timeshare group and antidepressant drugs melatonin can't.4. CMUS can bring Acrophase delaied, Rho and increased, Total Onset advanced.5.Decoction Jiawei Sini Decoction and melatonin treatment groups had significant antidepressant behavior effcet.6. Liver Decoction Jiawei Sini Decoction Mao Shi group and You Shi adminstration, mainly through adjustments gene circadian rhythm and content of gene to achieve antidepressant effects.
