The Effect Of Zuojin Pill On Expression Of The APC Gene And Avtivity Of The DNA Mehtyltransferase In Colorectal Adenoma

Research objectiveTo investigate the effect of Zuojin Pill on expression of the APC gene and activity of DNA methyltransferases (Dnmtl,Dmt3a and Dnmt3b) by the model of rat colorectal adenoma induced by 1,2-methylhydrazine,Thus to discuss the machanism of colorectal carcinoma developmet and anticancer effect of Zuo jin Pill.Research methods120 healthy wistar rats were randomly divided into three groups:the normal group,the zuojin pill group and the control group.We did nothing to the normal group rats,which of the zuojin group and control group were given a subcutaneous injection of DMH(25mg/kg once a week for 12 weeks) to establish a rat model of colorectal carcinomas.Meanwhile,rats of Zuojin group were given a medicine soup(i.g.1.4g/kg,once a day),After that, the control group were given a drinking of NaCL liquid (i.g.0.9% NaCL,once a day),Some of the rats in these three groups were killed randomly at the 11th,21th and 34th week after treated. Tumors which can be seen by our eyes were collected,Cut into slices,stained by HE and distinguished by pathological examination.Then,we detected expression of the APC gene in different tumor phases by the western blotting way and the activity of the Dnmt with the Elisa method.Research resultsThe relative expressin of APC is:normal group>zuojin group>control group,and the mount of each groups are greatly differentiated (P<0.01).However,it have nothing to do with Dukes staging.The Dnmt activity is:control group> zuojin group> normal group, all roups are significantly distinguished (P<0.05), and the activity of Dnmt1,Dnmt3b go up with the tumor immersion.Research conclusions The APC participate in the development of colorectal cancer and is an early event,which is independed of the phathology staging. Dnmt involvement in the occurrence of colorectal carcinomas and Dnmt1,Dnmt3b are corelated with colorectal tumor progression.Maybe,the rasing of APC expression and the decreasing of Dnmt activity have some connection with anticancer function of the Zuojin pill.
