Ulcerative colitis (Ulcerative colitic, UC) is an unknown etiology and pathogenesis of inflammatory bowel disease (inflammatoey bowel disease, IBD). Now that its incidence may be related to genetic, immune, infection and mental factors such as interactions, and on the pathogenesis of immunological research is currently more aspects.Antigen in human intestinal tract is in contact with the outside world the main area, so the local intestinal response to avoid the need to adjust the precision of food antigens and immune response to normal flora, while the effective defense of pathogens. In human and animal model studies have shown that regulatory T cells (regulatory T cell, Treg, Tr) in maintaining intestinal homeostasis play an important role. Reduction in the number of Treg, expression of surface molecules on defects, suppression of the damage associated with the occurrence of IBD. In recent years, Treg cells in the pathogenesis of IBD more and more attention.UC causes and the pathogenesis of disease complex caused certain difficulties to the treatment. Currently the treatment of ulcerative colitis are hormones, sulfa drugs and immunosuppressive agents, etc., but ineffective, and, side effects obvious. In recent years, Chinese people also seeking ways UC treatment, Chinese medicine is also more and more attention. Peony rhubarb soup for traditional Chinese medicine prescription, with heat detoxification, preliminary tests in our laboratory studies have shown that rhubarb Peony Decoction can improve symptoms of UC inflammation in mice, has a therapeutic effect, can improve the body's natural immunity, cellular immunity inhibited the hyperthyroidism, can inhibit cytokine TNF-a, IL-1βgeneration. Based on this experiment, this paper Peony Decoction of Rhubarb UC Treg cells in mice.ObjectiveUC model in mice to observe the rhubarb Peony Decoction on the treatment of UC, analysis and intestinal peripheral Treg cell number and function of peripheral blood cytokines, the pathogenesis of Treg in the changes of UC and Peony Decoction of Rhubarb Treg cells, further interpretation of rhubarb soup Peony UC pharmacological treatment of immune mechanisms.Method:First, the UC model in mice: selection of female BALB/c mice with 200μl,15mg/ml trinitrobenzene sulfonic acid (TNBS) in ethanol production by rectal administration of one mouse model of ulcerative colitis. Observe the process of weight loss occurs in mice, the highest disease index of 5.15±0.80, gross pathology score was 6.25±0.95. Compared with model group, group given rhubarb soup peony UC disease activity index and inflammation in mice significantly reduced the index (2.45±0.80,4.52±1.52) under the light microscope histological score was significantly lower (6.91±1.14).Model and the PPs in the peripheral blood Treg cell ratio significantly increased compared with normal group, treatment group than the number of Treg cells in model group, positive medicine group is lower than the number of Treg group, but both are higher than the normal group. Model group, serum IL-10, TGF-βwere significantly higher than the normal group, both treatment group were significantly higher than the model group, but also significantly higher than the normal group.Conclusion:Application of TNBS-Ethanol successfully established experimental mouse model of ulcerative colitis, this model modeling time is short, simple and practical, are acute onset, can be used as an animal model of UC pharmacology, efficacy of Rhubarb Peony Decoction can improve symptoms of UC inflammation in mice, mice UC has a therapeutic effect, can significantly reduce the peripheral blood, PPs and intestinal lamina propria in the number of Treg cells and increase in serum IL-10 and TGF-βlevels of cytokines, rhubarb soup can affect the peony and function of Treg cells is its treatment of experimental colitis in mice play a role in one of the mechanisms of inflammation.
