Objective:To study the effect of Angiotensinâ…¡inhibitors,which is angiotensin converting enzyme inhibitor (ACEI) benazepril and Angâ…¡1 receptor inhibitor (ARB) valsartan on Angâ…¡-induced cervical cancer cell line HeLa'antigen angiotensinâ…¡-1 receptor (AT1R), matrix metalloproteinase 2 (MMP2), matrix metalloproteinase 9(MMP9) protein and gene expression. To understand the major components of RAS system Angâ…¡and its receptor whether the role of AT1R and tumor invasion and metastasis factor MMP2, MMP9 are associated, the ACEI and ARB inhibitors can inhibit tumor cell metastasis is the role of factors for late cervical cancer in future clinical Transfer mechanism and treatment to help.Methods:The cervical cancer HeLa cells alone Angâ…¡, combined (ACEI) benazepril or ARB (valsartan), with the MTT cell proliferation to detect cell proliferation conditions, by immunofluorescence staining method to detect angiotensinâ…¡-1 receptor (AT1R), matrix metalloproteinase 2(MMP2), matrix metalloproteinase 9(MMP9) protein levels, Real-Time RT-PCR method of detection AT1R, MMP2, MMP9 gene relative expression. in the corresponding relative expression under the drug.Results:1. MTT method showed that Angâ…¡alone did not significantly promote cell proliferation, but the combined use of ACEI and ARB has inhibited the effect of Angâ…¡cervical cancer HeLa cell proliferation.2. Immunofluorescence staining results showed that:Angâ…¡significantly increased the use of cervical cancer HeLa cells angiotensinâ…¡-1 receptor (AT1R), matrix metalloproteinase 2(MMP2) and matrix metalloproteinase 9(MMP9)protein expression and AT1R protein expression and MMP2 and MMP9 correlated. But, once Angâ…¡conbine with ACEI and ARB inhibited the AT1R, MMP2 and MMP9 protein expression.3. Real-Time RT-PCR results showed that:Angâ…¡alone can significantly increase the cervical cancer HeLa cells AT1R, matrix metalloproteinase 2(MMP2) and matrix metalloproteinase 9(MMP9) expression of genes.And this AT1R gene expression related of MMP2 and MMP9 gene expression. But, if Angâ…¡conbine with ACEI and ARB inhibited the AT1R, MMP2 and MMP9 gene expression.Conclusion:1.The exogenous Angâ…¡through the AT1R to promote the cervical cancer HeLa cells matrix metalloproteinase 2(MMP2) and matrix metalloproteinase 9(MMP9) protein and gene expression, increasing the role of local RASS system may be one of the mechanisms of cervical cancer invasion and metastasis.2. Angâ…¡inhibitor ACEI (benazepril) and ARB (valsartan)can inhibit cervical cancer HeLa cells, matrix metalloproteinase 2(MMP2) and matrix metalloproteinase 9(MMP9)protein and gene expression. Inferred Angâ…¡inhibitors may become a new kind of inhibition of cancer metastasis and prolong survival of patients with cervical cancer drug.
