Objective:To investigate the expression levels of CD44,HIF-1αand other tumour makers in gastric cancer, to discuss the roles and significances in development of gastric carcinoma. Methods:Western-blot method was used to detect the expressions of CD44,HIF-1α,ENO1,PKM2,GAPDH,c-Myc,p53 and Bcl-2 in GES-1 and MKN-45 at 24h,48h and 72h under hypoxia (1% oxygen) conditions, compared to the control group under normoxic conditions (21% oxygen).Immunohistochemical method was used to examine the expressions of CD44,HIF-1a,EN01 and PKM2 in 63 samples of gastric carcinoma and corresponging to adjacent normal tissues. The statistic method was used to analyze the expression of CD44 and HIF-1a, as well as the relationship of gastric carcinoma clinicopathological parameters. Results:Under hypoxic conditions, the expression of CD44 was higher than control group in GES-1 and MKN-45 at 24h, with the extension of hypoxic culture time decreased. The expression of HIF-1α,ENO1,PKM2 and GAPDH were significantly higher than control group, with the extension of hypoxic culture time increased in MKN-45, but with the extension of hypoxic culture time the expression of ENO1,PKM2 and GAPDH were significantly lower in GES-1. The expression of c-Myc was significantly higher than control group, with the extension of hypoxic culture time, the protein levels slightly increased in MKN-45, opposite to GES-1. At hypoxic time point 24h, the expression of p53 was significantly higher than control group, but with the extension of hypoxic culture time increased, the protein levels significantly decreased. Immunohistochemistry method suggest that the positive expression rate of CD44 and HIF-1αwere 71.4%,61.9%respectively in gastric carcinoma and significantly higher than in adjacent normal tissues (15%,5% respectively). The expression of CD44 and HIF-1αwas not correlated with age,sex,tumor sites (P>0.05) in gastric cancer tissues, but the high expression of CD44 and HIF-1αwas significantly correlated with tumor size,differentiation degree,clinical stages and lymph node metastasis (P<0.05). The expression of CD44 was positivly correlated with HIF-1αexpression (P<0.05). Conclusions:CD44,HIF-1α,PKM2,ENO1,GAPDH,c-Myc,p53,E-ca and Bcl-2 were different levels of expression by hypoxia induced, they maybe together regulate gastric cancer cells to adapt hypoxia microenvironment and participated in gastric cancer carcinogenesis, but the mechanism need to more study. In the future, CD44,HIF-1αENO1 and PKM2 can be used as usefull markers for diagnosis and treatment in gastric carcinoma.
