| Objective:To study the effect of exogenous Apelin-13 on cardic function and the level of BNP and NO in plasma in rats with heart failure.Methods:In 70 male SD rats, initial body weight of which were weighed,10 were randomly collected as the control group, the rest of 60 ones were administered in intrapertoneal injection by adriamycin once every two days to build the model of heart failure. After the success of the model, the 60 rats were randomly divided into 3 groups:the heart failure model group (n=21), the Apelin-13 treatment group(n=16) and the Apelin-13+L-NAME intervention group(n=18). The Apelin-13 treatment group was treated by Apelin-13, the Apelin-13+L-NAME intervention group was intervened by L-NAME first, then by Apelin-13, all for one week. A week later, final body weight were weighed, left ventricular dysfunction was measured by intubation in the left ventricular; After the blood was taken out and the rats was executed, the heart weight was weighed; The level of BNP and NO in plasma was measured by ELASA; Histopathology of cardiac muscular tissue was obsevered in HE dyeing, proliferation of collagen fiber was observed in VG dyeing, and volume fraction of collagen fiber was measured too.Results:The death number in the heart failure groups was 11,6,8, and mortality was 52.38%, 37.50% and 44.44% respectively, difference between any two groups in the three groups had statistical significance ((P<0.05); After treating by Apelin-13 and compared with heart failure group, heart function parameters in the Apelin-13 treatment group could obviously be bettered, especially the maximum change rate of left ventricular pressure rise and fall, final body weigh increased, the level of BNP and NO in plasma and collagen volume fraction decreased, and histopathological changes of cardiac muscular tissue also bettered. Except the ratio of heate weight and final body weight, the difference of each parameter between any two groups in the four groups all had statistical significance ((P<0.05).Conclusions:Apelin-13 could obviously better cardic function, decrease the level of BNP and NO in plasma in rats with adriamycin-indeced heart failure, and might play myocardial action by the way of nitric oxide. |
