Objective:To investigate the efficacy of multi-frequency focused ultrasound (MfFU) in the treatment of alveolar hydatid disease in Kunming mice.Methods:Thirty mice infected subcutaneously with alveolar protoscolice were divided into 3 groups randomly and irradiated with different intensity of MfFU in vivo. The control group (group A) was not irradiated. All of the experimental groups were irradiated only once for 5 min. The irradiation power of the 3 transducers in the low-power group (group B) was 4W+4W+5W; That in the high-power group (group C) was 10W+10W+11W. After the irradiation, the morphological change of alveolar tissues was viewed with transmission electron microscope and light microscope; The survival rate of protoscoleces was evaluated with methylene blue staining; The mitochondrial content in the alveolar tissues was detected with laser confocal microscope; The Coomassie brilliant blue staining and succinate dehydrogenase (SDH) kit were applied for the measure of the amount of general proteins and the activity of succinate dehydrogenase of the alveolar echinococcus respectively.Results:After irradiated by MfFU, the cells on the germinal layer decreased. The mitochondrial swelled or broke. The endocytoplasmic reticulum showed swollen markedly. The karyotheca looked unclear. The microvillus shortened or disappeared. Compared with group A (82.33%), the survival rate of protoscoleces in group B (70.50%) and group C (59.83%) decreased significantly (P<0.05). The mitochondrial content of group B (105.46) were lower than group A (133.45)(P>0.05), while that of group C (70.90) much lower than group A (P<0.05); The amount of general proteins and the activity of SDH in both group B (3.07 mg/ml and 2.15 U/mg) and group C (2.87 mg/ml and 1.87 U/mg) were remarkably lower than those of group A(3.83 mg/ml and 3.50U/mg) (P<0.05).Conclusion:It is evident that the echinococcus multilocularis can be damaged by MfFU and the method has some curative effect. Objective:To establish a model of Swiss-albino mice infected with Hp, and to study the relationship between nickel and Hp.Methods:HP(SS1) and 10 clinical strains were orally inoculated to 80 mice of experimental group and NS were orally inoculated to 16 mice of control group respectively. Before the experiment, all mice were pretreated with antibiotics(ampicillin, gentamicin, azithromycin). At 1st and,2nd month after inoculation, stomach tissues of the experimental animal were used for the Hp colonization evaluation bacteriologically and molecular biologically.Results:In the all groups, no bacterial colonization was found, and after the PCR amplification of DNA extracted with gastric mucosa, the Vac A gene was not be expanded out, so the model experiment was failed.Conclusion:The reason of the failure maybe is that the strains used in the experiment is not susceptible to Swiss-albino mice.
