| Objective:To investigate the expression of TF mRNA in Bloodcell and TF antigen in plasma of the patients with arterious or venous thrombus in limbs.Methods:In order to undertake a case-control study,60 patients with thrombosis in limbs(case group) and 30 subjects without thrombosis(control group) in Fujian Province Hospital were studied.The case group were devided into two groups:arterious group and venous group of which all the patients were diagnosed by angiography.Basic clinical date such as AGE,SEX,Fasting Plasma Glucose(FPG),Triglyceride (TG),Total Cholesterol(TC),White blood-cell count(WBC),Neutroplil%(N%),Platelet count(PLT),Total Protein(TP),Albumin(ALB),Globulin(GLB),Alanine aminotransferase(ALT),Aspartate aminotransferase (AST),High density lipoprotein(HDL),Low density lipoprotein(LDL),Blood urea nitrogen(BUN),Serun creatinine(Scr) were collected. Detect the bloodcell TFmRNA of the patients with arterious or venous thrombus and control by RT-PCR, TF mRNA levels were scanned and semi-quantitative analyzed by Gel Imaging System. And detect TF level in plasma of the subjects investigated by means of ELISA. SPSS 13.0 was used to analyze all the data.Results:1.The levels of AGE,SEX,FPG,TG,TC,WBC,N%,PLT,TP,ALB,GLB,ALT,AST,HDL,LDL,BUN,Scr had no significant differences between case group and control group(P>0.05).2.Compared with the control group(0.24±0.09), arterious(0.43±0.10)and venous(0.39±0.11) thrombus groups are extremly higher in expression of the bloodcell TF mRNA,(P<0.05),the difference btween them is significative in statistics;while comparing arterious thrombus group (0.43±0.10) and venous thrombus group(0.39±0.11),we find that P=0.166,the difference btween them is insignificative.3.Compared with the control group, arterious and venous thrombus groups are extremly higher in expression of the TF level in plasma,(P<0.05),but there the difference btween arterious and venous thrombus groups is insignificative, (P>0.05).4.Pearson results showed that there was a positive correlation between bloodcell TF mRNA and plasma TF level in arterious and venous thrombosis group.Conclusions:1.TF mRNA expresses in human bloodcells.2.A high expression of bloodcell TF mRNA is found in patients with limb thrombus.3.Expression of TF in plasma is extremly high in patients with limb thrombus.4.There is a positive correlation btween the bloodcell TF mRNA and plasma level.5.The development of limb thrombus is correlated with the high expression of the bloodborn tissue factor. |
