[Objective] Mobilizing autologous endothelial progenitor cells with G-CSF and atorvastatin in UUO mice ,and comparing the differences on mobilizing autologous endothelial progenitor cell and affection on peritubular capillary between the two medicine.To discuss the differences of two medicine promoting kidney interstitial microvessel angiogenesis and provide the theoretical basis for using G-CSF and atorvastatin to delay the progress of Chronic kidney disease .[Methods]128 healthy female KM mice were randomly divided into 4 groups:①Sham operated group : mice was operated by sham surgery.②UUO group(unilateral ureteral obstructions group):mice was operated by ligation of right ureter.③U UO+G-CSF group :mice was operated by ligation of right ureter and injected subcutaneously of G-CSF50ug·kg-1·d-1.④UUO+ATO group:mice was operated by ligation of right ureter and received intragastric administration of atorvastatin 5mg.kg-1.d-1.8 mice of each groups were killed on the 7th, 14th, 21st and 28th day. The blood and renal tissue aliquot were collected .At each time point,Itams were detected as follows:①The percentages of CD133/VEGFR-2 double positive cells in peripheral blood mononuclear cells was evaluated by flow cytomerty.②Morphological changes of renal tissue were observed by HE and Masson stain and were investigated through light microscope. The method of renal interstitial fibrosis evaluation was used to evaluate the degree of renal fibrosis.③I mmuno-histochemistry method was performed to investigate the number of CD34 positive cells in order to evaluate the density of the peritubular capillary(PTC).[Results]The number of CD133/VEGFR2 double positive cells in peripheral blood reached its peak on the 7th day in each group,and then gradually decreased. The number of CD133/VEGFR2 double positive cells in UUO+G-CSF group had the most obvious increase rising about 6.5 times than the basic level.The UUO+ATO group up by about 4.8 times,the UUO group up by about 1.8 times and the Sham group up by about 1.3 times.The number of CD133/VEGFR2 double positive cells in Sham group falled to basic level and then remained steady on the 14th day.The number of CD133/VEGFR-2 double positive cells in UUO group droped to blow basic level on the 21th day.In this study period,the number of CD133/VEGFR-2 double positive cells in UUO+G-CSF and UUO+ATO groups was obviously higher than that in UUO group and Sham group(P<0.05).There was a tendency that the number of CD133/VEGFR-2 double positive cells in UUO+G-CSF groups was higher than that in UUO+ATO group.②P TC:The number of PTC in UUO,UUO+ATO,UUO+G-CSF groups decreased gradually after the surgery. there was no difference among groups on the 7th day.Compared to UUO group, the level of PTC was higher in UUO+G-CSF and UUO+ATO groups on the14th, 21st,28th days(P<0.05),but there was no difference between UUO+G-CSF group and UUO+ATO group.③R enal interstitial fibrosis evaluation:The degree of renal interstitial fibrosis evaluation in UUO,UUO+ATO,UUO+G-CSF groups increased in time-dependent manner.Sham group was normally,but there was no difference between UUO+G-CSF group and UUO+ATO group.④There was negative linear correlated with renal interstitial fibrosis degree and the density of PTC.[Conclusion]①G-CSF and atorvastatin induced the mobilization of endothelial progenitor cells from the bone marrow into the peripheral blood,and G-CSF was more effective than atorvastatin.②G-CSF and atorvastatin can delay the loss of peritubular capillaries in the mice with unilateral ureteral ubstruction.③There was no significantly difference on delaying the loss of peritubular capillaries and the progression of renal fibrosis between G-CSF and atorvastatin.
