AIM:To investigate HBVX's effect on expression of apoptosis factors.Methods:HBX gene eukaryon expression vector PCDNA3.1-HBVX was inject to mouse to establish mouse model for HBVX expression,mouse injected PCDNA3.1 and normal saline as control.The RT-PCR and Western blot were performed on mouse liver 48h after injection,and Semi-quantifed RT-PCR was used to evaluate the expression of Bax,Bcl-2,c-myc in 3 groups.Results:HBX mRNA was detected in injected mouse liver tissue by RT-PCR,expression of HBX protein was identified through Western blot,while no expression of HBX was detected in the normal control.Through Compared with normal control,expression of Bax(1.31±0.09vs0.91±0.11 P<0.05,1.31±0.09vs1.00±0.17 P<0.05),c-myc(1.6294±0.0672vs1.2869±0.0880 P<0.05,1.6294±0.0672vs0.9757±0.0397 P<0.05),Bcl-2(1.5567±0.1257vs0.6856±0.1554 P<0.05,1.5567±0.1257vs0.5488±0.1278 P<0.05)through Semi-quantifed RT-PCR were up-regulated.Conclusion:The mouse model haboring hepatitis B virus X gene is established successfully, HBVx can up-regulate either Bax,c-myc and down-regulate Bcl-2.
