Objective: To select genes of stress, heat shock protein and cell proliferation expressed differently in various stage of chemotherapy, then analyze the relationship of selected genes.Methods: Xenograft tumor models in nude mice were established by injection of human glioma cell line U251. Tumor-beared nude mice were treated with intraperitoneal injection of cisplatin (at a dose of 5mg/(kg.d) for 2-3 days ), control group are established before chemotherapy. Then chemotherapeutic and recurrent glioma model after chemotherapy were established. Gliomas were taken as the group after chemotherapy on the next day of chemotherapy; about 8 days after chemotherapy glioma were taken as the group of smallest glioma after chemotherapy; about 14 days after chemotherapy glioma were harvested as the group of recurrence after chemotherapy when gliomas grow to the size before chemotherapy. Expression of stress, cell proliferation and heat shock protein associated genes in pre-and post-chemotherapeutic gliomas were detected by oligo toxicology & drug resistance microarrays of Super Array(Catalog No:OHS-401). Data is analyzed by statistical software SPSS11.0.Results: Gene of stress, heat shock protein and cell proliferation expressed differently and time-relatedly. Apparent genes are as follows: (1) ABCC1 has a increasing expression after chemotherapy; (2) ATM and APC express increasingly in the diminish group, lower in the minimum group and increas again in the recurrence group; (3) HSPA6 and FGF2 express lowest in the diminish group and increased gradually in the next groups; (4) HSPB2 has the highest expression in the minimum group, the other groups express equally; (5) CCND1 express increasingly in diminish group, decline in the minimum and the recurrence group; (6) MT3 express decreasingly after chemotherapy. Cluster and correlation analysis show that there exist relationships of the genes talking above. HSPA6, HSPB2, CCND1 and FGF2 are positively correlated with ABCC1, MT3 is negatively correlated with ABCC1; HSPA6, CCND1 and FGF2 are positively correlated with ATM, HSPB2 is negatively correlated with ATM.Conclusion:Genes of stress,heat shock protein and cell proliferation have an different and time-related expression in different stage of chemotherapy. Apparent genes are as follows: ABCC1, ATM, HSPA6, HSPB2, APC, CCND1 and MT3. Gliomas may get their stress tolerance and recovery by these genes.
