| Objective By exploring the changes of NSE,GFAP,NCAM in hippocampus of young epilepsy rats induced by pentylenetetrazole(PTZ),and the effects of oxcarbazepine(OXC) on there expression, in order to investigate the nerve damage mechanism caused by epilepsy, and neuroprotective effects and its mechanism of OXC.Methods 50 male SD rats around 21 days of age were randomly divided into 5 groups: group A(negative control group),group B(positive control group),group C (low-dose OXC group),group D (medium-dose OXC group),group E(high-dose OXC group ) , and each group had 10 rats. Group A were intraperitoneal injected(IP) the equal amount of normal saline everyday; Group B were IP PTZ 40mg/kg .day, Group C~E were irrigate into stoach successively with OXC100mg/Kg,200mg/Kg,300mg/Kg before injected PTZ 40mg/kg·day. After twenty-one days, changes of body weight, behavioral performance were recorded, and the level of NSE,GFAP,NCAM and GFAPmRNA,GFAPmRNA in hippocampus were examined by immunohistochemisty and Real Time PCR.Results1.All epileptic rats had epileptic attacks in higher or lower degree,proving that the method to make rats epileptic model is successful. Compared with group B, latent time and degree of epileptic seizures induced by PTZ were respectively prolonged and alleviated in groups C~E(P<0.01).2. The growing of body weight of young rats in group E was slowly compared to each other group(P<0.01).3. Immunocytochemistry showed that there was nothing much immunopositive cells with NSE,GFAP and NCAM in hippocampus of young rats in group A,and the expression of them in hippocampus increased dramatically in group B(P<0.01,P<0.01,P<0.05), the expression of them were depressed in groups C~E (P<0.05,P<0.05,P<0.05). But there was no statistically significant differences in group D and E(P﹥0.05).The correlation analysis showed negative correlation between the expression of NSE,GFAP,NCAM and the dose of OXC(P<0.05,P<0.05,P<0.05).4. Real Time PCR indicated that the expression both of GFAPmRNA and NCAMmRNA in hippocampus of young rats in group A was much lower,and a dramatically increased was found in group B(P<0.01,P<0.05), the expression of them were depressed in groups C~E (P<0.05,P<0.05). But there was no statistically significant differences in group D and E(P﹥0.05). The correlation analysis showed negative correlation between the expression of GFAPmRNA,NCAMmRNA and the dose of OXC(P<0.05,P<0.05,P<0.05).Conclusion1. The modle of young rats of epilepsy induced by PTZ was a ideal model to explore Epilepsy activities .2. Neurons necrosis,the activity of astrocytes and synaptic reconstruction may play a important role in epilepsy .3. OXC could lessen the expression of NSE,GFAP and NCAM, inhibit neurons necrosis,astrogliosis and synaptic reconstruction,may have neuroprotective effcts against brain damage caused by PTZ kinding epilepsy in young rats.4. The neuroprotective effcts of OXC may relate to the dose of OXC. High dose of OXC may restrain the growing of young rats. |
