Objective To explore the effects of ulinastatin on SAP brain capillary permeability and its possible mechanismMethods The models of SAP were induced by retrograde injection of 5% sodium taurocholate into the bili-pancreatic duct.UTI group was made by UTI(10000IU/kg) venae femoralis injection within 5 minutes after SAP model was made.Sham operated(SO) group was made just by tipping the pancreas.Totally 48 helathy Sprague-Dawley(SD) rats were randomly divided into 3 groups:sham operation(SO) group, severe acute pancreatitis(SAP)group, ulinastatin(UTI) treated group. Each group was divided into two subgroups of 6h and 24h.There were 8 rats in each subgroup.The model was made by injecting 5% sodium taurocholate into pancreaticobiliary. The changes of TNF-α,pathohistology and water content were compared among different groups. The expression of occludin protein in brain was analyzed by immunohistochemistry method,and occludin mRNA level was measured by RT-PCR technique.Result TNF-α,pathohistology,water content ,occludin protein and mRNA level in group UTI were significantly diferent from those in group SO and SAP .Conclusions UTI could improve brain capillary permeability in severe acute pancreatitis rats. UTI may increase the expression of occludin protein by decreasing the level of TNF-α.
