| Objective:To investigate the protection and mechanism of Musk, Borneol and compatibility of them on cerebral ischemia, explain the resuscitation and awakening effect of compatibility of Musk and Borneol, and to explore the compatibility relationship between the two.Methods:①The animal models of cerebral anoxia and ischemia were established by sodium nitrite, guillotining,intravenous injection of magnesium chloride or air, ligation of carotid artery for 3 hours, observing the influence of the survival time and brain moisture content of the animal models after drugs, to To evaluate the protection of Musk, Borneol and compatibility of them on cerebral ischemia.②Measuring the activity of superoxide dismutase (SOD), content of malondialdehyde (MDA), vitality of Na+K+-Adenosine Triphosphate ase (Na+K+-ATPase) and Lactate dehydrogenase (LDH) by TBA method, Xanthine oxidase method, DNPH method and Pi method, exploring the mechanism of Musk, Borneol and compatibility of them on cerebral ischemia.Results: (1) Pharmacodynamic Study:Musk 66.67mg/kg, Borneo166.67mg/kg,133.33mg/kg and compatibility of them as 1:1,1:2 could remarkably lengthen the survival time of the mouse models which were established by sodium nitrite (P<0.05, P< 0.01).②All drugs could increase the times whose head had been broken, except Borneol 66.67mg/kg(P< 0.05或P<0.01). All drugs could increase the time of mouth opening of mice whose head had been broken (P< 0.05或P< 0.01).③Borneol 66.67mg/kg and the compatibility as 1:1 could prolong the time of mouth opening for breathing of mice injected with saturated magnesium chloride solution. (P< 0.05).④Musk 66.67mg/kg, Borneol66.67Mg/kg and the compatibility as 1:1 could prolong the time of mouth opening for breathing of mice injected with air (P<0.05或P<0.01).⑤All drugs could remarkably reduce the brain moisture content, except Borneol 200.00mg/kg and the compatibility as 1:3. (2) Biological mechanism:①All drugs could remarkably raise the activity of SOD, reduce the content of MDA of brain tissue (P<0.05或P< 0.01).②Borneol 200.00mg/kg and the compatibility as 1:3 could raise the activity of LDH of Brain tissue (P<0.05).③All drugs could remarkably raise the vitality of Na+K+-ATPase of Brain tissue (P< 0.05或P< 0.01)In the above experimental results,comparing with Musk and Borneol, the compatibility could not remarkably improve efficiency (P> 0.05)Conclusion:(1) Musk, Borneol and compatibility of them could protect brain because of anti-acute brain ischemia. Maybe this is the basis of pharmacodynamics of "resuscitation and awakening effect" by raise the activity of SOD, LDH, Na+-K+--ATPase and reduce the content of MDA of brain tissue. The mechanism could relation to Anti-free radical, Anti-metabolic acidosis and Improvement of energy metabolism.(2) The compatibility could keep the efficacy, whereas there is no greater efficiency of mutual reinforcement and mutual assistance on the compatibility than the single. |
