Objective:The aim of this study was to investigate the effect and mechanism of acupuncture on characteristic of locomotor activity circadian rhythm in Syrian hamster model of Chronic Fatigue syndrome.Methods:Use cold water swimming complex chronic restraint methods to copy Syrian hamster animal model of chronic fatigue syndrome. (1) use small animals recording and analyzing system to observe locomotor activity circadian rhythm in Syrian hamster model of Chronic Fatigue syndrome. The influence of activity circadian rhythm were measured by the changes of onset and acrophase phase of spontaneous activity.(3)Acupuncture were applied to "ganshu" (BL18), "pishu" (BL20) and "shenshu" (BL23) for 15min/d and lasted for 3 days during 5:00-7:00.(4)The expression of Per1mRNA and Per2mRNA were measured by RT-PCR method. The content of PER1 and PER2 were detected by western-blot method.Results:(1)The disorder of locomotor activity circadian rhythm was expressed by the model of CFS. The changes of model of CFS was delayed of onset and acrophase phase, reduced of activity. (2)Compared with Natural recovery group, the increased total number of locomotor activity could be observed in the acupuncture group. Acupunctured at Mao period could delay onset time and acrophase phase of spontaneous activity.(3)Compared with Natural recovery group,after acupunctured at Mao period, The expression of Per1mRNA and Per2mRNA and content of PER1 and PER2 were significantly increased.Conclusion:The disorder of locomotor activity circadian rhythm was showed in the model of CFS. Acupunctured at Mao period could delay onset time and acrophase phase of locomotor activity and significantly increased expression of Per1mRNA and Per2mRNA and content of PER1 and PER2 of CFS Syrian hamster.
