Cloning, Expression And Characterization Of Nanos Gene Of Schistosoma Japonicum

To clone and express Schistosoma japonicum Nanos like protein gene (SjNanos) and prepare its specific polyclonal antibody. Amplified SjNanos gene by PCR from Schistosoma cDNA library which was subcloned into pET28a(+) vector, then recombinant proteins were expressed in the presence of IPTG . The fusion protein was purified under denaturing conditions to immunize the rabbits with the polyacrylamide gel particles containing the recombinant protein for polyclonal antibody preparation. The sera were detected for antibody titer by ELISA assay and specificity by Western blot.SjNanos prokaryotic expression plasmid was successfully recombined and the target proteins with molecular weight 30ku were expressed with IPTG. The high titer(1∶128 000) polyclonal antibody was isolated from immunized rabbits and specifical protein band was detected by Western blot. SjNanos protein was successfully expressed and its specifical polyantibody was prepared, which would be useful for the further studies.
