Objective Establish chronic unpredictable mild mild stress mice model ,observe the effects of chronic unpredictable mild stress on mice ovarian reactive and oocyte development potential, and investigate if the local regulatory factor BDNF ,which related to oocytes development potential is involved in.Methods (1)Using 30 days of continuous stress,such as solitary raise, hunger, cold water swimming, etc, to establish chronic unpredictable mild stress mice model, appraisal the model by ethology, serology and brain tissue immunohistochemistry. (2) Observe ovulation situation after PMSG+HCG injection, in vitro oocytes culture to blastosphere stage after parthenogenesis activation, to observe the effect of chronic unpredictable mild stress on mice ovarian reactive and oocytes development potential. (3) Observe ultrastructure of oocytes and granular by perspective electron microscopy; observe the changes of BDNF expression level by immunohistochemica, to explore the possible mechanism of chronic unpredictable mild stress on mice ovarian reactive and oocytes development potential.Results (1) After 30 days of chronic unpredictable mild stress in mice, ethological detection shows that horizontal and vertical movement reduce obviously, serological detection shows that the level of corticosterone in mice rise clearly, immunohistochemistry shows that the positive neurons of corticotropin-releasing hormone in hypothalamus paraventricular nucleus increase.(2) Chronic unpredictable mild stress inhibite ovulation occurring, reduce Ovarian reactive,and influence oocytes developmental potential.(3) After chronic unpredictable mild stress,gap junctions are destroyed between oocytes with granular cells and more granular cell apoptosis, BDNF expression reduce in anteal follicle and Graafian follicle.Conclusions Mice are in stress condition and the activity of its HPA axis rise after chronic unpredictable mild stress. CUMS reduce the ovarian reactivity and oocytes development potential of mice. Chronic unpredictable mild stress destroyed gap junctions between oocytes with granular cells,increase granular cell apoptosis and lower the expression level of BDNF in anteal follicular and prevulatory follicle, suggest chronic unpredictable mild stress inhibit the expression of BDNF ,one of the ovarian autocrine and paracrine factors, then inhibit granular cell growth and proliferation and gap junctions between oocytes with granular cells in mice,thus affect the growth of follicles and oocytes developmental potential.
