Objective: To observe the effect of the Berberine (BBR) on the renal tubular epithelial cells (NRK-52E) of ischemis-reperfusion.Method: Using chemical hypoxia drug cobalt chloride (CoCl2) to stimulate NRK-52E cells to develop a chemical model of hypoxia-reoxygenation injury, with different concentrations (1×10-6,1×10-5,1×10-4mol·L-1) of the BBR dealing with NRK-52E cells. Using The Methylthiazolyldiphenyl-tetrazolium bromide (MTT) to test the activity of the cells, detecting the Malondialdehyde (MDA) and Superoxide Dismutase (SOD) content of each group, Hoechst33258 staining was used to detect the morphological changes of apoptotic cells of each group, Annexin V-FITC/PI staining was applied to detect the rate of apoptosis cell in each group.Result: MTT results showed that 1×10-4mol·L-1 BBR was toxic to the cells,the survival rate was 49.70%. The model group was 58.9%. and the survival rate of 1×10-6mol·L-1and 1×10-5mol·L-1 BBR were 85.0% and 67.30%. So it could increase the ischemia-reperfusion group cell survival, especially in 1×10-5mol·L-1 for the high. Hoechst33258 showed almost no apoptotic cells in the normal group, in the model group there were mostly nuclear staining of apoptotic cells, while BBR (1×10-5mol·L-1) pretreatment significantly reduced apoptosis cells. The SOD of the normal control groups was 24.33±0.15( 1×10-6U·L-1), the model group was 11.17±0.30(1×10-6U·L-1) and the 1×10-5,1×10-6mol·L-1BBR groups were 16.21±0.35 and 14.02±0.25(1×10-6U·L-1). The MDA of the normal control groups was 6.12±0.03(1×10-6mol·L-1), the model group was 15.81±0.25(1×10-6mol·L-1) and the 1×10-5,1×10-6mol·L-1BBR groups were 6.21±0.08 and 7.51±0.05(1×10-6mol·L-1). The MDA of ischemia-reperfusion group content was significantly higher (P<0.05), but the SOD was significantly lower (P<0.05). The pretreatment with BBR group, MDA was SOD significantly lower than before treatment (P<0.05), SOD was higher than the ischemia- reperfusion group (P <0.05), the role of BBR depends on the concentration between 1×10-6mol·L-1 and 1×10-5 mol·L-1. The Flow Cytometry showed that the apoptosis rate of the normal control group, 1×10-5, 1×10-6mol·L-1BBR group and ischemia-reperfusion group were 3.9%,11.1%, 32.2% and 56.2%, but the differences were significantly (P<0.05).Conclusion: The pretreatment with BBR on the ischemia-reperfusion cell has protective effects on renal tubular epithelial cells, especially in 1×10-5mol·L-1 was most effective. There may be two reasons. The BBR may be could increase SOD and decrease MDA, clean the oxygen-derived free radicals to alleviate oxidative stress. And the BBR may be could protect the damaged cells and inhibite the apoptosis. Objective:To study the therapeutic effects of berberine on renal function of Type II Diabetes Mellitus which contains SCr, BUN and 24hUALB.Methods:In clinicaltrial, a total of 60 patients with diabetes in our hospital were selected. They were randomly assigned to the treated group treated with berberine,and the control group treated with rosiglitazone, and the treatment lasted 3 months. The changes of SCr, BUN, 24hUALB, FPG, TG, CH, LDL-c, HbA1C and HOMA-IR were measured.Results: After 3 months, in the treated group the FPG, HbA1C, TG, CH, LDL-C, 24hUALB and HOMA-IR were decreased significantly(P<0.05). The SCr and BUN were also reduced, but the statistic difference was not found (P>0.05). The FPG, HbA1C, CH, TG, LDL-c, HOMA-IR and 24hUALB were decreased significantly(P<0.05)in the control group but the the statistic difference of SCr and BUN were not found (P>0.05).Conclusion: Berberine can decrease the 24hUALB of Type II Diabetes Mellitus. It maybe related with the FPG, TG, TC, LDL-C and HOMA-IR reduction. It suggests that berberine can prevent from the DN and it is worth studying it further.
