LY294002 on PI3K-Akt pathways and cells proliferation, apoptosis of anaplastic thyroid carcinoma cell line HTH-15. Methods: To perform the experiments, cells were divided into two groups:DMSO control group and LY294002(0~100μM) treated groups. The proliferation inhibitory rate of HTH-15 cells treated by different doses of LY294002 was evaluated by using MTT assay, The distribution of cell cycle was calculated by flow cytometry, And the protein expression level of p-Akt in HTH-15 cells were detected by Western Blot. Results: The growth of HTH-15 cell were inhibited by LY294002, and this inhibition was in a time- and dose- dependent manner. Treated with different concentration of LY294002, the percentage of cells in G1 phase was remarkably higher than that of DMSO control group(p<0.05); And the protein expression level of p-Akt in LY294002 group was significantly down-regulated compared to that of DMSO control group ( P<0.05) . Conclusion: PI3K-Akt pathway do exist in anaplastic thyroid carcinoma HTH-15 cells.LY294002 can inhibit the growth of HTH-15 cells through PI3K-Akt pathway, induces apoptosis. And PI3K-Akt pathway presents an appealing therapeutic target on Anaplastic. Objective: To investigate the effect and mechanism of LY294002 on cell invasion of anaplastic thyroid carcinoma cell line HTH-15. Methods: HTH-15 cells were divided into two groups , that is , A and B. The A group was control group without treatment. The B group received LY294002 (25,50μmol/L) for 24h.The protein expression of MMP-2 in HTH-15 cells were detected by Western Blot. And transwell chamber was applied to investigate cell invasion. Results: After the treatment of 25 and 50μmol/L LY294002 for 24 h, the protein expression of MMP-2 in LY294002 group was significantly down-regulated compared to that of DMSO control group ( P < 0. 05); And cell number penetrated the polycarbonate membrane under experiment groups was signification decrease compare with DMSO control group(p<0.05),the cell invasion capacity was also inhibited when treated with LY294002. Conclusion: LY294002 can inhibit the MMP-2 express in HTH-15 cells through PI3K-Akt pathway, and reduces cell invasion ability.
