Objective:To detect the expression of the Neuropilin1 in the human gastric carcinomas; To investigate Neuropilin1 (NRP1) on the growth of nude mice transplanted gastric carcinoma by constructing small interfering RNA (SiRNA).Methods:The expression of NRP1 in the human gastric carcinomas as well as the normal gastric tissue was detected by immunohistochemistry. Microvessel density (MVD) was detected by marking vascular endothelial cells with CD34; The hairpin SiRNA-NRP1 sequence targeting to human NRP1 gene was designed to synthesize a pair of complementary oligonucleotides. The oligonucleotides was annealed and cloned to plasmid of pGenesil-1.1. The recombinant plasmid was identified using enzyme digestion analysis and DNA sequencing. Gastric cancer cell line SGC7901 was injected subcutaneously to the right hip of nude mice to construct transplanted model of gastric carcinoma; The recombinant plasmid was injected to the right hip of nude mice after neoplasia formation. General condition of nude mice was observed and the weight as well as the volume of the nude mice transplanted tumor was measured. The growth of tumor tissue and angiogenesis was observed by pathological section .The expression of NRP1 and microvessel density in the nude mice transplanted tumor was observed by immunohistochemistry.Results:The expression of NRP1 in the human gastric carcinomas was higher than that of normal gastric tissue(P<0.05). Enzyme digestion analysis and DNA sequencing showed that the target gene had been cloned to plasmid of pGenesil-1.1. The weight and volume of the nude mice transplanted tumor in interference group was lower than that of control(P<0.05). pathological section show that tumour cell and tumor vessel density in interference group was lower than that of control. The immunohistochemistry showed that the expression of NRP1 and microvessel density in the interference group was lower than that of control(P<0.05)Conclusions:The expression of NRP1 was significantly correlated with MVD in the human gastric carcinomas; The SiRNA-NRP1 could inhibit the expression of NRP1 so as to curb the growth of tumor angiogenesis in the nude mice transplanted gastric carcinoma ,which finally helped to inhibit the growth of tumor.
