Background:The tumor is refers in various carcinogenic factor influence, resulting in loss of cells in the gene level on the growth of normal control, resulting in abnormal proliferation of cell clones formed.As one of a large class of cancer, is a major cause of death of one of its local invasion and distant metastasis, with the important features is its most important reason to cause death.Tumor suppressor gene (tumor suppressor gene) is a large class can inhibit cancer cell division and inhibit the function of the gene.Normal cells in the body of a negative regulatory role of proliferation genes, when the tumor suppressor gene deletion, inactivation or aberrant expression, they tend to promote the growth of malignant cells showed a loss of control.Oncogene is the induction of its gene in cancer cells and as inherent gene in the human or animal cells controls cell growth, is considered a negative expression of tumor suppressor genes or dominant oncogene expression may be caused by abnormal cell proliferative or cancer of the main adjustment mechanism.Therefore, the abnormal expression of tumor suppressor gene and oncogene expression may be the main reason of the development of cancer.PTEN(phosphatase and tensin homology deleted on chromosome ten,PTEN) as a dual phosphatase tumor suppressor gene, located on chromosome 10q23.In a variety of cancer research, it shows that the segment loss of heterozygosity occurs frequently, a nonsense mutation, missense mutation and so forth may be the reasons of the occurrence and development of tumor, such as: gliomas, thyroid cancer, breast cancer, oral squamous cell carcinoma and so on.In the studies have shown that, PTEN gene in normal tissues showed high expression, in tumor tissue with low expression or loss of expression . Therefore, PTEN may be one of the important factors in the occurence and development mechanism of cancer .Currently, the research on adenoid cystic carcinoma of salivary gland is small, so further study of salivary adenoid cystic carcinoma of the existence of abnormal expression of PTEN gene, the existence of other similar low expression of the tumor, whether the expression of PTEN protein relate to PTEN gene transcription, it becomes necessary.Objective:The experiment apply RT-PCR, western bloting method to study differences between the normal salivary gland tissue and salivary adenoid cystic carcinoma of the expression of PTEN gene and protein .Study on whether the PTEN gene relate to the occrence of adenoid cystic carcinoma of salivary glands ,and whether PTEN protein expression and PTEN gene is associated RNA reverse .In order to provide a relevant experimental evidence of relativity between the salivary gland tumors and tumor suppressor gene PTEN gene , and for the laboratory diagnosis of salivary gland tumors providing experimental evidence and theoretical basis.Materials and Methods:1, Collecting pathological diagnosis specifically for the patients with adenoid cystic carcinoma of salivary adenoid cystic carcinoma and corresponding normal tissue, according to the instructions of RNA to extract total RNA , reverse line PCR, PCR amplification products by 1.5% agar sugar gel electrophoresis gel images using Bio-Rad imager observations, analysis of normal salivary gland tissue and salivary gland adenoid cystic carcinoma in PTEN gene expression.2, In the salivary gland adenoid cystic carcinoma, PTEN protein shows low expression. in normal salivary gland tissue ,expression of PTEN protein in cancer tissue was higher than the expression of state.According to Bio-Rad gel imager images of X-ray imaging for quantitative determination of the gray value, using SPSS13.0 statistical software ,and statistical analysis test: salivary adenoid cystic carcinoma of the expression of PTEN protein significantly lower than normal salivary gland, and a statistically significant difference between the two (P <0.05), at the same time, PTEN protein expression in both the PTEN gene has a certain relevance. Results:1.the expression of PTEN RNA in salivary gland adenoid cystic carcinoma tissues is lower expression, the expression of PTEN RNA in normal salivary gland organization more highly than cancerous tissue. According to the Bio -Rad gel image earners picture grayscale value quantitatively determined, using SPSS13.0 statistical software by the statistical analysis: t test.the expression of PTEN RNA in salivary glands adenoid cystic carcinoma tissues significantly lower than normal salivary gland organization, and the differences between them are statistically significant (P < 0.05).2. the salivary gland adenoid cystic carcinoma tissues PTEN protein expression is lower expression, The expression of PTEN protein in normal salivary gland tissues a cancerous tissue more highly than cancerous tissue.. According to the Bio -Rad gel image earners of grey value of X-ray, quantitatively determined by SPSS13.0 statistical software by t-test for statistical analysis: salivary gland adenoid cystic carcinoma tissues PTEN protein expression significantly lower than normal salivary gland organization, and the differences between them are statistically significant (P < 0.05), meanwhile, the expression in both PTEN protein PTEN gene expression with have certain correlation.Conclusions:The expressions of PTEN genes exist both in adenoid cystic carcinoma and normal . In normal salivary gland tissue ,expression of PTEN gene was highly expressed state, in the salivary gland adenoid cystic carcinoma of PTEN gene expression was significantly lower ,in normal cells and salivary gland salivary gland adenoid cystic carcinoma cell line (ACC-2), the expression of PTEN protein of a corresponding, the difference of PTEN gene expression level in a certain extent, reflect the PTEN gene in adenoid cystic carcinoma play a role in the process.PTEN gene as a more specific phosphatase activity of tumor suppressor genes in tumor occurrence and development mechanism, plays an important role , may be used as early diagnosis and monitoring of adenoid cystic carcinoma markers for the early detection to improve the treatment efficiency, and improve quality of life of patients, provide better services for patients.
