Objective:In order to establish an ideal molecular detection method which was covering a variety of viruses, rapid, high sensitivity and specificity.We compared Seeplex multiplex RT-PCR and VRDAL multiple nested PCR to detect the same 164 nasopharyngeal swabs collected from children aged <3-year old hospitalized due to acute respiratory infection during 2009 to 2010 in Shanghai.The more appropriate method can reflects the true distribution of the virus lineages in the children.Methods:From May 2009 to July 2010, 164 nasopharyngeal swabs were collected from patients under 3 years of age with fever and ARI symptoms consulting the Paediatric Department of Xinhua Hospital in Shanghai, China. The cDNA synthesis was performed using extracted viral nucleic acids. VRDAL multiplex nested PCR for eight common respiratory viruses were performed with specimens;then,the Seeplex(?) RV15 ACE Detection kit for the detection of 15 different virals were tested with the same specimens.The two multiplex PCRs were compared and we analysised of the characteristics of respiratory viral infections in 0-3 age group children during 2009-2010 in Shanghai, its predominant viral pathogens, relationship of virus detection and season, age, gender, to provide guidance for clinical prevention and treatment of respiratory tract infections.Results:1. The experiment proved that Seeplex multiplex RT-PCR was reliable and sensitive, specifically amplified 15 different respiratory viruses target gene.2. Seeplex multiplex RT-PCR detected causative viruses in 129 patients (78.7%),including 49 co-infected The first three regnant virus were hRV, IVA, PIV-2.Conclusions:1. Two multiplex PCRs compared showed Seeplex multiplex RT-PCR was superior to VRDAL Multiple Nested PCR in total virus and co-infection virus detection and it can be widely used in clinical and research laboratories.2. In this study, using Seeplex multiplex RT-PCR method we successful studied on the infect profiles of respiratory viruses in children under three-year age and layed the foundation in research and development high-throughput detection methods of micro-organisms.
