Gardenia is for both food and traditional Chinese medicine in China, geniposide is the most abundant in gardenia and plays an important role in the medicinal effects of gardenia. In some countries, geniposide is used in folk medicine. For therapy , geniposide showed anti-tumor , anti-oxidative and so on.Microglia has been considered to represent the brain's internal immune system.Activated microglia produce some compounds like reactive nitrogen species (RNS), inflammatory factor, reactive oxygen species (ROS), and any other neurovirulence factors.These factors may injury neurons,then result in neurodegenerative diseases.Further research of microglia activities may offer a significance approach in probing the new therapeutic direction in preventing and treating PD and AD.The purpose of this topic clarify the inhibitory mechanism of geniposide in LPS-stimulated murine microglia N9 cells.This issue proved that geniposide attenuated LPS-induced activation of murine microglia N9 cells and inhibited over-production of NO and iNOS in N9 cells.In addition, Geniposide suppressed the formation of intracellular ROS in LPS-induced murine microglia N9 cells. Geniposide showed blocking effect on the phosphorylation of p38 MAPK, ERK1/2. Geniposide also inhibited the drop-off of IκB from the NF-κB.Then inhibited the NF-κB translocation from cytoplasm into nucleus of N9 cells.
