In recent years,target therapies for a variety of tumors have achieved clinically significant response.The rationale for this treatment is to detect some targeted molecule in the tumor.Such detection not only provide important data for the program in therapeutic,but also providing relevant information for prognosis.It was reported in recent years that there was abnormal expression about hedgehog signaling pathway in gastric cancer,and it was involved in the development of gastric cancer,which could be seen in many human tumors. More and more evidence revealed that EMT is an early important process in invision and matastasis of many tumors.Many studies have shown that hedgehog signaling pathway may be related to the invision and metastasis of gastric cancer.But the research on which the mechanism of hedgehog signaling pathway caused the invision and metastasis of gastric cancer is still little.Therefore, the paper will focus on the relation between hedgehog signaling pathway and epithelial-mesenchymal transition, and further study the mechanism of invision and metastasis in gastric cancer.Therefore, we selected the key factors which were upstream factor Shh and downstream factor Gli1 in hedgehog signaling pathway,and typical fctors Snail and E-cadherin in epithelial-mesenchymal transition.We detected firstly threir expression in 54 cases of gastric cancer tissues,their adjacent tissues and 30 cases of non-cancerous gastric mucosa tissues by immunohistchemistry. And then in vitro experiment, we would futher analysised interreaction the interaction between hedgehog signaling pathway and epithelial-mesenchymal transition.Objective: The purpose of this study is to investigate interaction between Hedgehog signaling pathway and epithelial-mesenchymal transition in invasion and metastasis of gastric cancer.Methods: The protein expressions of Shh,Gli1,Snail and E-cad in 54 cases of GC tissues,their adjacent tissues and 30 cases of non-cancerous gastric mucosa tissues were detected by immunohistochemisty, and the correlation with clinicopathological features was analyzed. The human gastric carcinoma SGC-7901 cells was parted to the control group,empty vector group,cyclopamine interference group,Snail siRNA group and combination interference group with cyclopamine which is the special blocker of Hh signaling pathway and Snail siRNA. The expressions of Gli1,Snail,E-cadherin mRNA in each group were detected by useing RT-PCR. The effect of interference factor on invasion and metastasis ability about gastric carcinoma cells each group was detected by transwell small room. The effect of interference factor on heterogenous adhesion ability about gastric carcinoma cells was detected by adhesion experiment.Result: The positive rates of Shh,Gli1,Snail and E-cad were 68.52%, 61.11%, 75.93% and 25.93% in GC tissues, respectively. Those were 27.78%,24.07%,40.74% and 70.37% in adjacent tissues respectively. Those were 36.67%, 26.67%, 30% and 96.67% in non-cancerous gastric mucosa tissues respectively, with statistically significant between gastric cancer tissues and adjacent tissues(P<0.05).The expression of these four proteins was related to the differentiation and lymph node metastasis(P<0.05).The expression of Shh, Gli1 and Snail was positively correlated with each other (P<0.05), but they were negatively correlated with E-cadherin (P<0.05). The Gli1 mRNA expression in the cyclopamine group and combination group was more weaker than that in the control group,empty vector group and Snail siRNA group (P<0.05). The expression of Snail mRNA in the cyclopamine interference group,Snail siRNA group and combination interference group was more weaker than that in the control group and empty vector group (P<0.05) ,while E-cadherin mRNA was more stronger (P<0.05).There were less cells invading through the Matrigel in the cyclopamine interference group,Snail siRNA group and combination interference group than that in the control group and empty vector group ,and so do the heterogenous adhesion ability of gastric cancer cells (P<0.05).Conclussion: In the course of the development of gastric cancer,hedgehog signaling pathway may be involved in regulation of Snail to low the expression of E-cadherin ,which result in epithelial-mesenchymal transition ,and thus promoting the invasion and metasasis of ganstric cancer.Experimental results of combined detection of these four antibodies can be used as molecular markers for clinical detection. This will help to assess whether it was invasion and matastasis of gastric cancer, and make personalization treatment about gastric cancer and prognosis for reference.
