Objective:To explore the correlation between circulation tumor cell and lymph node metastasis, in order to evaluate the value of CK19 mRNA and EGFR mRNA for the clinical diagnosis of laryngeal carcinoma micrometastasis.Methods: From the 40 nude mice choose 35 as experimental group,and then divided the 35nude mice into 5 groups randomly,7 mice in each group in order to established the laryngeal cancer nude mice into tumor model,the rest 5 nude mice as control group.The mice in the experimental group were killed in 2,4,6,8 and 10 weeks after injection respectively.The expression of CK19 and EGFR mRNA in the peripheral blood and tumor tissue were detected by RT-PCR assay. The expression of CK19 and EGFR in tumor tissue by immunohistochemistry and lymph node use continuous pathological dyeing confirmed whether the transfer.Results:Nude mice model transplanted with human laryngeal carinoma was successfully established. There were 20 nude mice development into lymph node metastasis,and the positive transfer rate was 57.1%(20/35).None of CK19 and EGFR mRNA were detected in peripheral blood from control group,CK19 mRNA-positive rate was 54.3%(19/35)and 80%(28/35)respectively in peripheral blood and tumor tissue in the experimental group,EGFR mRNA-positive rate was 37.1%(13/35) and 74.3%(26/35)respectively in peripheral blood and tumor tissue in the experimental group. CK19 and EGFR was the specificity tumor markers for the CTC, the lymphatic metastasis was related to the CTC(P<0.001), and the metastasis was often happened in the exponential growth phase of the tumor. The immunohistochemistry found that the protein positive expression rate of CK19 and EGFR were 80%(28/35) and 85.7%(30/35) in tumor tissue respectively..Conclusions:The expression of CK19 and EGFR mRNA in the peripheral blood could contribute to provide predictive information of lymphatic metastasis.And the CK19 and EGFR was the specificity tumor markers for the CTC, indicated that there were correlation between CTC and lymph node metastasis in laryngeal carcinoma.
