| ObjectiveTo reform the traditional soft tissue expander. With it , the analogue skin was constructed in vivo and evaluate its feasibility in order to provide experimental basis for clinical application.Methods1. Reconstruction of conventional tissue expander:The conventional tissue expander of single injection pot will transformed into double injection pot,The original injection pot turned towards inner dilated balloon and the additional injection pot turned towards outer dilated balloon with 10ml volume of dilated balloon. The designed model was elaborated by Winner Plastic Surgery limited company in Shanghai.2. Cultivation of rabbit keratinocytes :The abbit keratinocytes received primary culture and were purified and amplified, making preparation for anaphase of cell implantation and the further experiments.3. Construction of analogue composite skin:Each of ten new-zealand albino rabbits was subcutaneously embedded with new tissue expander at the back of them. fibrous cyst emerged around the expander and primary autoallergic keratinocytes suspension was injected by the additional injection pot after 1w, that is to say, rabbit keratinocytes were implanted into the lacune between fibrous cyst and expander to construct analogue composite skin.4. Constructed effectiveness of analogue composite skin:Ten new-zealand albino rabbits were included and randomly allocated into two groups, including 1w (Group A) group and 2w (Group B) group. the expander was removed ,and the fibrous cyst around the expander was examined grossly and histologicaly to evaluate the epithelialization on the fibrous cyst surface.Results1. In both groups, all experimental animals survived and the wound was healed at the first rate.2. rabbit keratinocytes were cultured and purified and amplified successfully.3.By gross examination, the fibrous cysts in group A and group B were not significantly different. But, it is quite diferent between two groups. In group A, epithelial islands but not integrity epithelial layer appeared on the surface of the fibrous cyst.In group B, integrity epithelial layer was formed that meaned epithelialization was completed. Fibrous layer was closely connected with epithelial lamina and then developed integrated skin similar to skin structure.ConclusionIt is possible to construct analogue skin in vivo with the reformed expander, that play important role in the experiment study. |
