Objective: To validate the suppression of colchicines in the formation of hypertrophic scar so as to provide the experiment foundation and theory support that the colchicine is effective and safe in the treatment of proliferative scar, stable and redundant rabbit ear hypertrophic scar model was induced and four examination target were applied.Materials and Methods: Sixteen healthy adult New Zealand rabbits were divided randomly into four groups: group A(saline),group B (colchicines , 0.15mg?ml),group C (colchicines , 0.30mg?ml) and group D(colchicines , 0.60mg?ml ). Six wound surfaces in size of l cm×l cm were created down to bare cartilage on the ventral surface of each ear. 50μl saline and colchicines in different concentrations were injected into the wounds in the 7d, 14d, 21d, 28d, 35d, 42d, 49d and 56d after the surgery. Two rabbits were sacrificed in each group in the 28d and 56d. Then①hypertrophic index(HI) was evaluated,②the morphological changes of scars were observed by Hematoxylin and Eosin (HE)dying,③the quantity of hydroxyproline in the hypertrophic scar was measured by the chloramine T method and④the expressions of matrix metalloproteinase-l(MMP-1) and tissue inhibitor of metalloproteinase-1(TIMP-1) were determined by immunohistochemistry in the three experimental groups and the normal control group in the two time points respectively.Results: HI was higher, the dermis layer was thinner, the quantity of fibroblast was reduced, the arrangement of collagenous fibre was more regular and the expressions of MMP-1 and TIMP-1 was lower in the experimental groups compared to the normal control group, while the quantity of hydroxyproline in the hypertrophic scar was lower in the experimental groups compared to the normal control group(P<0.05).Conclusion: the suppression of colchicines in the formation of hypertrophic scar is obvious,maybe through inhibiting the proliferation of fibroblast and decreasing the contents of collagen fibers.
