Objective To observe the expression of IFN-γ, IL-4, IL-13 in hepatic fibrosis rats and rats treated with TGF-β1siRNA, TIMP-1 siRNA and TIMP-2 siRNA, explore the changes of cytokines in siRNAs antifibrotic.Methods The liver tissue which collected from liver fibrosis rats before and after treatment of TGF-β1siRNA, TIMP-1 siRNA and TIMP-2 siRNA were the research objects. According to the different interference target genes, the liver tissue were divided into two group A (including normal group, model group, negative group and the TGF-β1siRNA treatment group) and group B(including normal group, model group, negative group, the TIMP-1 siRNA treatment group and the TIMP-2 siRNA treatment group). The liver tissue stored in formalin was embedded in paraffin for the detection of IFN-γ, IL-4 and IL-13 by immunohistochemistry. The liver tissue stored in liquid nitrogen was for the detection of IFN-γ, IL-4 and IL-13 by Western blot and Real-time PCR.Result The expression of Th1 cytokines IFN-γin the model group and negative group were significantly lower than the normal group, the expression of Th2 cytokines IL-4, IL-13 were significantly higher than the normal group (P <0.05), either group A or group B. The expression of IFN-γ in the TGF-β1siRNA treatment group were significantly higher than the model group, while the expression of IL-4, IL-13 were no significant difference comparison to the model group and negative control group (P> 0.05). The expression of IFN-γin the TIMP-1 siRNA treatment group and the TIMP-2 siRNA treatment group were significantly higher than the model group, and also the expression of IL-4, IL-13 were significantly lower than model group(P <0.05).Conclusions There were higher expression of Th2 cytokines IL-4, IL-13 and lower expression of Th1 cytokines IFN-γin liver fibrosis rats. This phenomenon proved the existence of Th1/Th2 paradigm in liver fibrosis rats. All TGF-β1 siRNA, TIMP-1 siRNA and TIMP-2 siRNA had certain therapeutic effect on liver fibrosis. But after interference the expression of TGF-β1, IL-4 and IL-13 decreased unconspicuous. Contacting with the biological characteristics of IL-4 and IL-13, it was not conducive to the recovery of liver fibrosis. After interference the expression of TIMP-1 and TIMP-2, the expression of IL-4 and IL-13 were also decreased. It inhibited the pro-fibrotic of Th2 cytokine, and conductive to anti-fibrosis treatment.
