House fly (Musca domestica) distributes all over the world, is regard as one of the most important disease vector. Insecticides have been extensively used in China for the management of public health pest. The current control strategy of disease vector is based primarily on the application of insecticides. House fly have regarded as one of the most important disease vector but its resistance to insecticides have been found in many province. However, insecticides which were excessively appled has caused the occurrence of house fly resistance. Organic phosphorus (OP) and Pyrethroid had extensively been used because they were low toxic to the mammalian and rapid knockdown effect on house fly. The resistance monitoring to house flies is essential to guide the rational use of insecticides and resistance management.1. The wild resistnace of house flies from nine urban garbage dumps in the province of Bejiing, Tinajin, Guangdong, Shanghai, Sandong and Jilin to OP (Malathion and Diazinon) Permethrin and Deltmaethrin were evaluated in 2010. The bioassay results showed that the housefly resistance to Malathion has reached a higher level. LD50 value of TTY strain was 280.81 (219.85-377.51)μg/fly, and its resistance ratios is 305-fold. The resistance of the houseflies in all area to diazinon was not found.2. The resistance of house flies to four insecticides, malathion, diazinon, permethrin and deltmaethrin were screened. The resistance of other three strains were significantly risen besides ZYH-DI after selecting 4 generations.3. A 500bp fragment sequence by the amplification of the allele specific method had been gotten. Then the genotype of ZYH F0 generation with a digestion was detected. Three genotypes, homozygous sensitive (SS), heterozygous (RS) and homozygous resistant genotypes (RR) were found. The genotype frequencies of sensitive homozygous (SS), heterozygous (RS) and resistant homozygous (RR) were 0.43%,7.45% and 2.18%, respectively.4. Six genotypes (L/L,L/F,F/F,L/H,H/H,F/H) had been detected by the synthesis of the primers and allele specific amplification (AS-PCR).
