Torque teno virus (TTV) was first discovered in 1997 in Japan, from the serum of a patient with acute post-transfusion hepatitis of unknown etiology by Nishizawa, using the method of RDA.TTV is a new hepatitis virus after the discover of A-G hepatitis virus, but it was classified into the family Anelloviridae. TTV is a small non-enveloped single-stranded circular DNA virus, the genome of it has a range of 2.1-3.9kb in length and contains two main open reading frames (ORFs), ORF1 encodes the structural protein, ORF2 main encodes the non-structural protein. However, in addition to humans, TTV can also be detected from non-human primates, porcine, avian, bovine and ovine, eline and canine, camels, sea lions, sea turtle and so on. The lengths have significant deviation in different genegroups or species, human TTV have at least 40 genotypes. For the wide host scopes, the many transmission route and the relationship with other diseases, many scholars pay close attention to TTV.Based on the sequence of SAa-10(AB060594), primers were designed to detect the TTV of the genotype SAa-10.In the study, a total of 355 samples were detected (150 children fecal samples from hospitable children aged from 1month to 8 years, in Shanghai; 109 serum in Shanghai and 96 fecal samples from Inner Mongolia).The nucleotide homology of the positive samples sequences and the AB060594 are all nearly 90%. This confirms that genetic variability rather than geographical variance exists among TTV's in infected human.The complete genome sequences of one isolate was amplified by nest PCR, and the phylogenetic tree was constructed by Mega 4.0,the result shows that genetic variability rather than geographical variance exists among TTV's in infected human.Using the Protean of DNA-Star, the epitope in ORF1 was predicted and the region full of epitope was amplified to legate to the vector PET-30.The result shows that the protein could express effectively and exist in cytorrhyctes, which to do a significant exploration of researching TTV' ELISA and its vaccine antigen.
