Objective:To test the expression of a-SMA protein in Human Umbilical Vein Endothelial Cells (HUVECs), which stimulated by arecoline, as the concentration of arecoline and intervention time increased. And further to gain the possible pathogenesis of OSF.Methods:With different concentrations (5,10,20,40,80μg/ml) of arecoline stimulate HUVECs, respectively, after 1 day (A group),4 days (B group),7 days (C group),10 days (D group), detected the expression of a-SMA in HUVECs using the method of immunocytochemistry.RESULTS:(1) After the HUVECs stimulated by various concentrations of arecoline in one day (A group),4 days (B),7 days (C group), there were no a-SMA expression detected in each group by immunocytochemistry; (2) the expression of a-SMA protein were detected by immunocytochemistry after the HUVECs stimulated by arecoline for 10 days (D group) (P<0.05).Conclusions:Exposure of HUVECs to Arecoline can induce endothelial/mesenchymal transdifferentiation, suggesting that endothelial /mesenchymal transdifferentiation may play an important role in the pathogenesis of OSF. Objective:To investigate the effect of Hyperbaric Oxygen (HBO) to the expression of a-SMA in Human Umbilical Vein Endothelial Cells (HUVECs), which occurred the changes of endothelial-to-mesenchymal transition by stimulated with arecoline, in order to find out the possible mechanism of Hyperbaric Oxygen therapy to OSF.Methods:After the human umbilical vein endothelial cells intervented by different concentrations (0,5,10,20,40,80μg/ml) of arecoline for 10 days, the cells were seeded in 24-well plates, divided into the hyperbaric oxygen group (0.2MPa Hyperbaric Oxygen 2 times/day for 2 days),control group in atmospheric pressure air and the control group without treatment, detected the expression of a-SMA by using immunocytochemistry.Results:(1) Between hyperbaric oxygen group and the atmospheric air group, there were no significantly differents in the expression levels of a-SMA protein among groups intervented by different concentrations of arecoline (P> 0.05); (2) Hyperbaric oxygen group compared with the untreated control group, the expression levels of a-SMA protein were significantly decreased among groups (P<0.05); (3) Compared atmospheric air group with control group before treatment, the expression levels of a-SMA protein were significantly decreased among groups(P<0.05).Conclusion:(1) Hyperbaric Oxygen therapy may have no direct influence on the expression of a-SMA were decreased in human umbilical vein endothelial cells; (2) Stopping the intervention of arecoline, the expression of a-SMA in human umbilical vein endothelial cells may be lost and become normal as the passage of the cells.
