Objective To study the activation of epithelial growth factor receptor (EGFR)expression in astrocytes,and explore the relationship betweem EGFR and the astrocyte'sactivation.Methods Astrocytes were cultured purely from newborn rats,and were divided into threegroups: the control group which was cultured with the normal medium, the activation groupwas cultured with the medium which has 20ng/ml ciliary neurotrophic factor (CNTF) inthe normal medium ,and the inhibit group is that was cultured with the medium 30umol/lGenistein in the normal medium after astrocytes were activitied . The shape and thenumber of astrocytes were examined by immunocytochemistry,the expression of glialfibrillary acidic protein (GFAP) and EGFR were examined by RT-PCR.Results The activation group compared with the control group, the number ofastrocytes cells was increased,cell bodies become larger,processes were longer,and woveninto a mesh,astrocytes were activited,the expression of GFAP and EGFR increased.andwhen Genistein was applied in the activitied astrocyte,the astrocytes's number was nomore increased and cell body become smaller,processes were reduced,astrocyte'sactivation was inhibited,and the expression of GFAP was decreased.Conclusion 1.The expression of EGFR was increased after astrocytes were activited;2.Genistein can inhibit the astrocyte's activation by EGFR.
