| Objective: To study the temporal patterns and expression sense of brain-derived neurotrophicfactor (BNDF) and cycloxygenase-2 (COX-2) and platelet-derived growthfactor-B(PDGF-B)in rats with closed traumatic brain injury(TBI) Methods: Based on Marmarou's model of injury, eighty-five Wistar rats both sexes, were randomly divided into normal control group(n=5),sham group(n=5), postmortem injury group(n=5) and experimental group(n=45) [it included 5 mices in every phase(lh,3h,6h,12h,1d,3d, 7d,14d)]. With the model of closed traumatic brain injury, the expression of DBNF, COX2 and PDGF-B was study by immuohistochemical method (SP) in brain of Wistar rats in control group with different time after injury. And the morphological changes of the rat brain were obserbed by light microscope. The immunostaining results were meassured with statistics analysis. Results: The expression of BDNF increased sharply at 1h after injury, compared with the nomal group were different(P<0.05), peaked at 3h,decreased and returned to basal level at 7d. At 1h after injury; After closed traumatic brain injury for 1-2h, the expression of COX-2 in brain tissue increases progressively, compared with the normal group were different(P<0.05). and reaches to the peak level after 12h, and return to the normal largely after 5d; The expression of PDGF-B enhanced, compared with the normal group were different(P<0.05). and marked up-regulation were seen at 3-5d,then they decreased but remained above control till 14 day after injury.There was statistical significance on experimental brain injury group and postmortem brain injury group(P<0.05).Conclusion:1)It can be used to authenticate lifetime damage and postmortem brain injury by checking out the expression of BDNF, COX2 and PDGF-B.2) Closed traumatic brain injury induced expression of BNDF, COX2, PDGF-B. The regular level change of them with injury time might provide evidence for detecting brain injury. |
