Objective:To investigate the interleukin-1A+4845, interleukin-1B-31 (IL-1A+4845. IL-1B-31) genotype and their association with the susceptibility of chronic periodontitis and coronary heart disease, analysis the relationship of gene polymorphism and the detection of Prevotella intermedia of atherosclerotic plaques, and introduce the new quota technology simultaneously for detecting the quantity of prevotella intermedia from subgingival plaques and atherosclerotic plaques. Methods:DNA was obtained from buccal swabs of 127 subjects with moderate to severe chronic periodontitis,123 subjects with coronary heart disease,87 subjects with moderate to severe chronic periodontitis and coronary heart disease and 117 matched healthy control individuals Genotypes of IL-1A+4845/Fnu 4H1 and IL-1B-31/Alu I were analyzed by PCR-RFLP method. Then the differences in distribution of genotype were compared. DNA was obtained from subgingival plaques and atherosclerotic plaques of 38 subjects with chronic periodontitis and coronary heart disease. Then the prevotella intermedia were detected by using the polymerase chain reaction and its relationship with the allele T of interleukin-1B-31 (IL-1B-31) were observed. Comparing the quantity of prevotella intermedia from subgingival plaques and atherosclerotic plaques by real-time qPCR technology. Results:1) There was no statistical differences about the distribution of IL-1A+4845 genotype among the four groups.2) A significant statistical difference about distribution of IL-1B-31 genotype was found among the four groups. A more significant over represen-tation of IL-1B-31 allele 2 was found in CP group than the healthy control individuals, and there was no statistical difference.3) In coronary atherosclerotic plaques sample from the 38 patients, Prevotella intermedia (Pi,42.1%) was detected, in subgingival plaques, Prevotella intermedia (Pi,68.4%) was detected, the concordant presence of the same periodontal bacteria DNA in subgingival plaques and in coronary atherosclerotic plaques in the same patient was Pi 9 (29.0%).4) The allele T in the subgingival plaques which examed Pi are much higher than the non-examined one, and this difference has the statistical significant.5) The design of primers in real-time qPCR has good specificity and sensitivity. Conclusion:This result suggests that the allele of IL-1B-31 may be associated with CP susceptibility. The presence of Prevotella intermedia DNA in coronary atherosclerotic plaques could indicate that periodontal bacteria may play a role in the coronary heart disease process.
