Objective:1. To observe the expression of hypoxia-inducible factor la (HIF-la) in vitreous body in the patients with proliferative diabetic retinopathy (PDR),proliferative vitreoretinopathy (PVR) and the control group and to analyze its changing regulation.2. To analyze the influencing factors about the express of HIF-1αin the patients with proliferative vitreo-retinal diseases.3. To discuss the role of HIF-la in the pathogenesis of proliferative vitreo-retinal diseasesMethods:The vitreous samples were collected during the vitrectomy from 71 eyes of 69 patients (experimental group) including 39 eyes with proliferative diabetic retinopathy (PDR) and 32 eyes with proliferative vitreoretinopathy (PVR). There were 8 vitreous fluid samples in the normal control group. The concentrations of HIF-1αin vitreous body were detected by enzyme-linked immunosorbent assay (ELISA).Results:1. The concentrations of HIF-1 a in vitreous body in the eyes of PDR, PVR and the normal control group were (301.63±23.18). (284.88±24.42),(16.38±3.56) mg/L respectively, showing a significant difference among the three groups (F=273.72, P=0.00). The concentrations of HIF-la in vitreous body were obviously increased in the eyes of PDR and PVR group in comparison with the normal control group respectively (t=23.32, P=0.00;t=21.74, P=0.00 respectively). The concentrations of HIF-1αin vitreous body were increased in the eyes of PDR group in comparison with PVR group (t=3.01, P=0.00).2. For the patients with PDR, the concentrations of HIF-1αin vitreous body were (274.58±30.05),(298.37±17.34),(315.94±18.30) mg/L atⅣ,Ⅴ,Ⅵstage respectively, showing a significant difference among the three stages (F=8.87, P=0.00). The concentrations of HIF-1αin vitreous body were obviously increased at the V stage and the VI stage in comparison with the IV stage respectively (t=2.44, P=0.02;t=4.08, P=0.00 respectively). The concentrations of HIF-1αin vitreous body were increased at the VI stage in comparison with the V stage (t=2.59, P=0.01).3. For the patients with PVR, the concentrations of HIF-1αin vitreous body were (267.44±22.45),(295.34±19.33)mg/L at C, D stage respectively. The concentrations of HIF-1αin vitreous body were increased at the D stage in comparison with the C stage (t=3.72, P= 0.00).4. For the patients with proliferative vitreo-retinal diseases, the HIF-1αconcentration was (302.69±21.65) mg/L in vitreous with nephropathy and (292.50±25.44) mg/L without nephropathy, and there was no significantly statistic differrnce (P=0.22).5. For the patients with proliferative vitreo-retinal diseases, the HIF-1αconcentration was (293.54±28.03) mg/L in vitreous with hypertention and (294.40±23.32) mg/L without hypertention, and there was no significantly statistic differrnce (P=0.89).6. The linear regression analysis between the concentrations of HIF-1αin vitreous body in the eyes of proliferative vitreo-retinal diseases and the course of ophthalmopathy was made and the regression equation was Y=1.096X+286.613. There was inferior correlation between the concentrations of HIF-1αin vitreous body in the eyes of experimental group and the course of ophthalmopathy.7. For the patients with proliferative vitreo-retinal diseases, there was no correlation between the sex, the age and HIF-la concentration in vitreous (r=-0.207, P=0.08; r=0.016, P=0.89 respectively). There was no correlation between fasting blood glucose (FBG), duration of DM and HIF-1αconcentration in vitreous (r=-0.159, P=0.33; r=0.012, P=0.94 respectively).Conclusion:1. Intravitreous HIF-1αlevel in the patients with proliferative vitreo-retinal diseases was higher than the control group.The more severe degree of fundus pathological changes was, the higher level of intravitreous HIF-1αwas in the patients.2. HIF-1αmay play an important role in the pathogenesis of proliferative vitreo-retinal diseases.
