Objective:1.To test the expressions of IDO,Foxp3,E-CD,MMP-9,CD 105 in human gastric carcinoma;2.To evaluate the relationship between above mentioned proteins and various clinicopathology factors;3.To analyse the correation between Foxp3,E-CD. MMP-9,CD 105 and IDO;4.To analyse the relationship IDO with tumor metastasis infiltrating and immunological function, and further understand the role of IDO in tumorigenesis of gastric carcinoma, and further understand the relationship in IDO role in the development of, and explore its relationship with prognosis.Methods:The clinicopathologic data of 40 advanced gastric carcinoma patients were analyzed retrospectively.All patients underwent radical gastrectomy in Tianjin Medical University in January 2002 to December 2006.①To analyze the clinical pathological data for all patients;②To analyze the relationship between prognosis and its pathological types, lymph nodes metastasis, the 5-year survival rate, et al;③The expression of IDO,Foxp3,E-CD,MMP-9,CD 105 were determined by immunohistochemical staining of tissue from gasric cancer specimens, peficancerous and lymph nodes; and evaluated the relationship between expression of above mentioned proteins and various clinicopathological factors;④To analyse the correation between Foxp3,E-CD,MMP-9,CD105 and IDO.Results:1.In 40 cases of stomach specimens, IDO positive expression rate and cell index were 45%(18/40) and 22.03±19.48%, respectively. Foxp3 positive expression rate and cell index were 82.5%(33/40) and 2.28±1.74%, respectively. The E-CD low expression rate was [67.5%(27/40)], The MMP-9 expression rate was [77.5% (31/40)], MVD of CD 105 markers was 28.45±12.60.2. Significant association of IDO expression in gastric carcinoma tissues with lymph node metastasis and TNM staging (P<0.05) were found; Significant association of Foxp3 expression in gastric carcinoma tissues with lymph node metastasis (P<0.05) was found; Significant association of E-CD expression in gastric carcinoma tissues with tumor biggest diameter and pathologic types were found (P<0.05); Significant association of MMP-9 expression in gastric carcinoma tissues with TNM staging was found (P<0.05); Significant association of CD105 expression in gastric carcinoma tissues with tumor biggest diameter, T stage and TNM staging were found (P<0.05).3. IDO correlates positively with Foxp3 significantly with regard to positive expression rate (r=0.417, P<0.05) and positive cells index (r=0.599 P<0.05), linear regression equation was y=1.097+0.054 x (y:Foxp3 positive cells index, x: IDO positive cells index), no significant association of IDO expression in gastric carcinoma tissues with E-CD,MMP-9,CD105 was found (P>0.05).4. Univariate analysis revealed that T staging, lymph node metastasis, TNM staging, IDO expression, CD 105 expression were prognostic factors of stomach cancer after radical gastrectomy. Multivariate analysis revealed that T staging,TNM staging, IDO expression were independent prognostic factors of stomach cancer after radical gastrectomy(P<0.05).Conclusion:1. IDO,Foxp3,MMP-9,CD105 were over-expressed in human gastric carcinoma, E-CD shows low expression.2. IDO correlates positively with Treg significantly, suggesting that IDO contribute to metastasis of gastric carcinoma by inducing differentiation of Treg possibly.3. IDO expression was independent prognostic factor of stomach cancer after radical gastrectomy.
