ObjectiveThis study was to observe the effects of PI3K inhibitor LY294002 on PI3K/Akt signal pathway, cell growth, cell invasion and cell apoptosis in human gastric adenocarcinoma SGC-7901 cells.MethodsHuman gastric carcinoma SGC-7901 cells were treated with LY294002, and western blot was used to measure the expression of proteins in the PI3K pathway including P-AKT,MMP-2,MMP-9,Bcl-2 and CyclinD1. Proliferation,invasion and apoptosis were detected by MTT,Flow Cytometry and Transwell assay.ResultsCompared with SGC-7901 cells in the control group and DMSO group LY294002 significantly downregulated the expression of P-AKT,MMP-2,MMP-9,Bcl-2 and CylinDl. Proliferative activity was suppressed in the cells treated with LY294002 starting on the second day. The results of the Transwell assay showed that the number of cells travelling through the matrigel was 81.0±2.56 in the control group, 81.3±1.52 in the DMSO group, and 44.6±2.52 in the LY294002 group.There were statistical differences among the three groups (F=255.1,P=0.000). The flow cytometry results showed that there was G0/G1 arrest in the LY294002 group. The percentage of apoptotic cells was increased when SGC-7901 cells were treated with LY294002ConclusionLY294002 inhibits the PI3K/AKT signal transduction pathway.
