| ObjectiveTo observe the effects of breviscapine on airway remodeling in asthmatic rats and explore the possible mechanisms.MethodsForty SD rats were randomly divided into 4 groups(10 in each): normal group, model group, dexamethasone group, breviscapine group. The asthmatic models of rats were established by sensitized and challenged with OVA and Al(OH)3. Hematoxylin-eosin (HE) staining was used to observe the pathologic histology of lung tissue. Samples of hydroxyproline was measured with alkaline hydrolysis. Matrix metalloproteinase-9(MMP-9) and tissue inhibitor of metallo- proteinase-1(TIMP-1) in lung tissue was detected with immunohistochemical techniques. The expression of MMP-9 and TIMP-1 in lung tissue was measured with western-blot.ResultsCompared with normal group, the inflammatory cells in airway, smooth muscle were severe in model group. Statistic analysis showed that the hydroxylproline, MMP-9 and TIMP-1 in lung tissue were higher in model group than those in normal group(P<0.05). Those changes in dexamethasone group and breviscapine group were lower than those in model group(P<0.05). But its between the two groups was not significant(P>0.05).ConclusionsBreviscapine could reduce the recruitment of inflammatory cells and the formation of hydroxyproline causing collagen deposition, inhibit the expression of MMP-9 and TIMP-1 in tissue of lung, Indicating that the therapeutic mechanism of breviscapine for airway remodeling is related to reduce airway inflammation, the formation of hydroxyproline and regulate dynamic balance MMP-9?TIMP-1. |
