Detecting The Effect Of Huluan Decoction On The FoxP3's Expressions Of Immunity POF Mouse Model With The Western-blot Method

Objective:Explore the mechanism of Huluan Decoction on POF through detecting the effect of Huluan Decoction on The FoxP3's expressions of immunological POF model mice with the Western-blot method.Methods:The immunological POF model mice were set by BALB/C. The Laboratory mice are divided into six groups, respectively blank group, model group, conjugated estrogens Tablets group and Huluan Decoction of high, medium and low doses group. Then observe the sexual cycle changes in mice. After 60 days executed mice, take out the ovarian tissue, weigh and computational ovarian index, and do Morphological observation on ovarian tissue with microscope in the Western-blot way to detect the FoxP3's expressions of immunological POF model mice.Results:1.The comparison of Model group with Blank group, the ovarian weight index decrease obviously, and there is a significant difference(P<0.01).In model group, the FoxP3's expression decrease obviously, too.(P<0.01). 2. The each treatment group which is compared with model group, The ovarian weight index and the FoxP3's expression of Huluan Decoction of high doses group and conjugated estrogens Tablets group increase obviously (P<0.01). But on Huluan Decoction of medium group and low doses group,there is no obvious difference (P>0.05).3. The comparison of Huluan Decoction groups with conjugated estrogens Tablets group, The ovarian weight index and the FoxP3's expression of Huluan Decoction of high doses group have no obvious difference (P>0.05), But there is a significant difference on Huluan Decoction of medium group and low doses group (P<0.01).4. The comparison of three Huluan Decoction groups. There are significant differences statistically in comparison between Huluan Decoction of high doses group and the other two groups, (P<0.01).And there is no obvious difference between Huluan Decoction of medium group and low doses group (P>0.05).Conclusion:Huluan Decoction has the improved effect on ovaritis and ovarian failure induced by autoimmune reaction. Huluan Decoction can remarkably increase the index of ovary weight and vital follicle of ovary. The lymphocyte and plasma cell infiltration decrease. The sexual cycle is regular and the estrous cycle is prolonged. The expressions of FoxP3 and the number of CD4+CD25+Treg cells all increase to inhibit the injury of autoimmune injury.
