Effects Of QWBZP On Anti-virus Protein Expression In The Intestinal IEL CD3~+T Cell Nuclear Of HRV Infection Suckling Mice

Object:To investigate effects of QWBZP on anti-virus protein expression in intestinal iEL CD3+T nuclear of HRV infection suckling mice, then to further elucidate its anti-HRV mechanism.Method:1. HRV suspension was used to infect 5 days old suckling mice through mouth, and after suckling mice feces was confirmed positive for rotavirus antigen, the diarrhea model of HRV-infected suckling mice was established.2. The suckling mice were randomly divided into model group (MG); Qiweibaizhu Powder group (BG); virazole group (ZG); inhibitor group (YG); Qiweibaizhu Powder plus inhibitor group (BYG) and virazole plus inhibitor group(ZYG), and set a group without HRV-infected as control group (NG); The mice of each treatment were instilled the corresponding oral drug (50μl each time) respectively through mouth, while the mice of NG and MG were given the equal cell fluid.3. (1)CD3+T cell were separated from the small intestinal mucosa in 15min, 1h,2h,6h,12h,24h,32h, and 48h after treatment, and the expression of p-JAK1 and p-STAT1 protein in CD3+T cell cytoplasm were observed by western blotting. (2) CD3+T cell were separated from the small intestinal mucosa in 32h,40h and 48h after treatment, and the expression of PKR, p-elF 2a,OAS and RnaseL protein in CD3+T cell nucleus were observed by western blotting.(3) 5 days after drug treatment, feces were obtained and diluted gradedly, then cultured in MA 104 cell to observe the effects of drug treatment on virus clearance by immunofluorescence.Results:1. Expression of p-JAKl in BG was increased, reached the climax during 6h and 12h, after 6h, expression of p-JAK1 protein in BG was higher than those in MG, BYG, ZG, ZYG and YG at the same time (P<0.05).2. Expression of p-STAT1 in BG was significantly higher than those in MG, NG, BYG, ZYG and YG (P<0.05) at the same time.3. Expression of PKR, p-eIF 2a, OAS and RnaseL protein in BG changed over time, increased after 32h, reached the climax during 40h and 48h, and then decreased after 48h, with the expression higher than those in Mg, NG, BYG, ZYG and ZG (P<0.05) at the same time.4.5 days after treatment, there was no significance in HRV clearance rate in feces between BG and ZG.Conclusion:Qiweibaizhu Powder eliminate HRV in intestine of the HRV infected sucking mice, possibly through activating p-JAKl and p-STAT1 in intestine iEL CD3+T cell cytoplasm, and promoting the expression of Antiviral protein in intestine iEL CD3+T cell nucleus.
