Objective:A sophisticated regulatory procedures or 'biological clock' is ubiquitous in every species of life, it govern to turn on or off the auxesis, division and death of cells. The sophisticated regulatory procedures or 'biological clock' is known as cell cycle regulation mechanism. It is under the control of relevant genes, performed in a specified sequence and in accordance with specific rules to regulate the auxesis, division and death of cells. We know, the pathogenesis and development of the tumour are directly bound up with failure of cell cycle control. Wee1 is one of important kinase of cell cycle, the activation or inactivation plays a significant role in process of cell cycle, furthermore, the activity of Wee1 closely contact with phosphorylation. This article disusses the expression and the relationship of Wee1 and phosphorylated Wee1 proteins in colorectal cancer of different periods. To investigate the relativity of three proteins and the correlation between the clinical types, pathological stage and metastasis. Meanwhile, it also explore whether Weel and p-Weel can be novel hallmarks for clinical patients. In order to enhance the diagnostic rate of digestive tumors, and to provide the experimental basis for the tumors of early diagnosis, early treatment and prognosis.Methods:This study applies immunohistochemistry two step method, detect the expression of three proteins (Wee1B11, p-Wee1Ser3 and p-Wee1Ser642) in the tissues of colorectal cancer and normal tissues. The 100 fresh samples of the carcinoma coming from 100 patients with colorectal cancers who did not undertake chems and radiotherapy before the operation, and the samples are confirmed by pathology.52 fresh samples are normal colorectal tissues, which are 10cm far away from the tumor. and the correlation among them were studied, using statistic methods such as the chi-square (χ2) test and Spearman correlation test. P value less than 0.05 is considered to have statistic significance, and P value less than 0.01 is considered to be significantly different.Results:The postive rates of Wee1B11, p-Wee1Ser53 and p-Wee1Ser642 in carcinoma of colorectal are at a higher rate,68.00%,85.00% and 91.00% respectively. However the postive of the three proteins in normal colorectal tissues are at a lower rate,9.62%, 21.15% and 42.31% respectively. The positive staining for Wee1B11 and p-Wee1Ser53 are observed in nucleolus of tumor cells mainly, and little positive staining in the cytoplasm. The positive staining for p-Wee1Ser642 is observed in cytoplasm of tumor cells mainly, and little positive staining in the nucleolus. The statistical results reveal the postive rates in carcinoma of large intestine tissues for the three proteins are significanly superiority to the one in normal intestine tissues (P<0.05). However, the expression of Wee1, p-Wee1Ser53 and p-Wee1Ser642 in carcinoma have no relations with the patient's age, gender and size of tumors (P>0.05), and have positive correlation with tumor differentiations (P<0.05), have negative correlation with invasion of adventitia (P<0.01), lymph metabasis (P<0.05), distant metabasis (P<0.05) or Dukes stages (P<0.05). The statistical results with the Spearman correlation test reveal that there is significanly positive corrleation between the three proteins (P<0.05).Conclusions:1.The study detects the Wee1B11, p-Wee1Ser53 and p-Wee1Ser642 in the carcinoma of colorectal cancer for the first time. There are almost no Wee1B11, p-Wee1Ser53 and p-WeelSer642 expressions in the normal normal mucosa tissues, but the expressions of Wee1 and p-Wee1 are significantly higher in carcinoma of colorectal. 2.The expressions of Wee1 and p-Wee1 are related to the tumor differentiation and prognosis, and are related to the invasion of adventitia, lymph metabasis, distant metabasis and Dukes stages.4.It's a possibility that during the emergence and development of colorectal cancer, the ability of 'G2/M checkpoint" repairing damaged DNA is abated by the disorder of activation or inactivation of Wee1 and phosphorylated Wee1, and then leading to the disorder of cell cycle regulation mechanism.4.To revealing the mechanism of Wee1 and p-Wee1 in colorectal cancer, and offering new evidences for the early diagnosis, early treatment and prognosis of colorectal cancer, we should pay more attention to the cytology and molecular biology deeply.
