[Objective] To establish the serum proteins diagnosis model and identify serum protein biomarkers specific for Primary Biliary Cirrhosis form healthy people with surface enhanced laser desorption/ionization time—of flight masss pectrometry (SELDI—TOF—MS),we try to detect and identify the special protein in order to find the sensitive and specific experimental date for diagnosis of PBC.[Methods] The sera of 21 cases of patients with Primary Biliary Cirrhosis and 20 cases of healthy people were test by weak cationic exchanger (WCX2) and surface enhanced laser desorption/ionization time—of flight masss pectrometry of Ciphergen Company. Proteomic fingerprinting of serum were identified and analyzed by Biomarker Wizard Software and Biomarker Patterns Software. Software.SPSS 13.0 will be used for statistical analysis. Then the diagnosis model will be established with BPS 5.0. A decision tree model that represented the high sensitivity and specificity for PBC patients from healthy controls to find the abnormal serum protein biomarkers specific then PBC will be identified.[Results](1) The most discriminative mass/charge (m/z) values serving as potential biomarkers for PBC were identified in range,1000-50000Da. From among all candidates.There are 153 protein peaks with statistical difference (p<0.05) including in 15 protein peaks from Kolmogorov-Smirnov Test and the 3 protein peaks from Mann-Whithey u.(2) Construction of the decision tree classification algorithm was performed by Biomarker Patterns Software,5 discriminative peaks were generated.The following peaks were:m/z values of 23356.2Da,9589.96 Da,5860.40,8583.71 Da,11924.5 Da, the decision tree could differentiate PBC and healthy people with the sencitvity of 95.000% and specificity 90.476%.[Conclusion] (1)We can establish high sencitvity and specificity diagnosisi model of PBC patients serum by using SELDI—TOF—MS.
