Objectives:To research the expression of the D2-40 and the CCR7 in the non-small cell lung cancer tissues; to explore the expression of the D2-40 and the CCR7 in the non-small cell lung cancer and the lymph node metastasis along with its characteristics of clinical pathology; to provide some theoretical bases for the clinical diagnosis and the treatment of NSCLC and estimating the risk of lymphatic metastasis; to search a new target and to provide new ideas and methods which may be used to treat lung cancer.Methods:1,Via application of the MAX Vision immunohistochemical staining technique,the expression of the lymphatic endothelial markers——D2-40 was measured in the 104 cases non-small cell lung cancer and the 5 cases of normal lung tissues and 10 cases peficancerous tissues of non-small cell lung cancer,analysed and observed and located the lymphatic vessels in the light microscope.2,By means of application of the MAX Vision immunohistochemical staining technique, the expression of the CCR7 was measured in the 104 cases non-small cell lung cancer and the 5 cases of normal lung tissues and the 10 cases peficancerous tissues of non-small cell lung cancer.3,To proceed statistic analysis about the the expression of the D2-40 and the CCR7 in the non-small cell lung cancer tissues and the related clinical pathological factors.Results:The lymphatic vessels marked by D2-40 were rarely seen in the intratumoral tissues of the non-small cell lung cancer.The lymphatic vessels mainly distributed in the peritumoral tissues of the Non-small cell lung cancer. We found the MLVD counts of the non-small cell lung cancer were obviously more than that of the normal lung tissues (7.81±2.22>4.20±1.07,P<0.001).The MLVD counts of the 10 cases peficancerous tissues of non-small cell lung cancer is 4.9±1.46.The MLVD counts of the large cell carcinoma,the adenosquamors carcinoma,the squamous carcinoma,the adenocarcinoma are respectively 7.0±3.79; 8.60±2.38; 7.99±2.08; 7.51±2.08.2,The MLVD counts of the group of the 47 cases lymph node-positive patients is obviously more than that of the group of the 57 cases lymph node-negative patients (8.39±2.35>7.33±2.00,P<0.05).3,CCR7 is 82.7% positive (86/104) in the 104 cases non-small cell lung cancer and CCR7 is 20% positive (2/10) in the 10 cases pericancerous tissues and CCR7 is 20% positive (1/5) in the 5 cases normal lung tissues.The CCR7 positive rate of the large cell carcinoma,the adeno-squamous carcinoma,the squamous carcinoma,the adenocarcinoma are counted up to 83.3%, 77.8%,82.1%,84.0%.4,The CCR7 was 87.2% positive(41/47)in the group of the 47 cases lymph node-positive patients and 61.4% positive (35/57)in the group of the 57 cases lymph node-negative patients (Chi-square test:P<0.05,χ2=87.4).5,The expression of the MLVD marked by D2-40 and the CCR7 is positively related(coefficient of correlation,r=0.639, P<0.000).The MLVD counts in the group of the CCR7-positive(8.51±2.03) is more than that of the group of the CCR7-negative(6.01±1.59).The difference was of statistical significance (P<0.05).6,The expression of the CCR7 and the MLVD counts have nothing to do with these patients'smoking status; age; T-stages.They are connected with lymph node metastasis,but it is no sense expressing difference between pNl and pN2.Conclusion:1,The D2-40 has specificity and sensitivity to lymphatic endothelial cells,and it can make a distinction between lymphatic vessels and vascular vessels,and can mark clearly tumor lymphatic vessels.Tumor lymphatic vessels mainly distributed in the peritumoral tissues of the Non-small cell lung cancer.Which can apply to research about the tumor lymphatic vessels.2,The positive rate of CCR7 and the MLVD marked by D2-40 in the NSCLC are more than the group of normal lung tissues.3,The MLVD and theCCR7 are positively related with lymphatic metastasis.In the NSCLC,the positive rate of CCR7 of the lymph node-positie group is more than that of the lymph node-negative group.4,The CCR7 and the MLVD is also positively related. In the NSCLC,the MLVD of the CCR7-positie group is more than that of the CCR7-negative group.5,The MLVD and the CCR7 have nothing to do with smoking status,age,T-stages.They are connected with lymph node metastasis(pN- and pN+),but it is no sense expressing difference between pNl and pN2 on statistics and clinical meanings.In non-small cell lung cancer,in the lymph node metastasis positive-group,the expression of the CCR7 is positively related to the MLVD marked by D2-40.The D2-40 staining only presents in the lymphatic endothelial cells;the Cancer cell highly express CCR7;as result,which is early events of lymph node metastases.The detection of the CCR7 along with the D2-40 are expected to as inchoate and more effective signals to judgment lymph node metastases.It might provide strong theoretical bases for the treatment NSCLC and the lymphatic metastasis.
