| Polyhydroxy alkaloids (PHAs) of a-glucosidase inhibitor are a class of compounds in which the oxygen atom of carbohydrate ring is replaced by a nitrogen atom. PHAs could competitively inhibit a-glucosidase because of a structural resemblance to the sugar moiety of the natural substrate. Therefore, the peak blood sugar content after meal was significantly decreased by using the a-glucosidase inhibitor. This effect is therapeutic benefit to diabetes. Morus L. is the main source plant of the PHAs, and 1-deoxynojirimycin (DNJ) is the highest content compound in it. DNJ not only has therapeutic potential application in diabetes but also plays roles in anticancer and antivirus. It could make value in health care and medicinal use because of its efficiency and low toxicity. There are variety species of mulberry trees in China and they are wide distribution. Thus, in order to make good use of DNJ in mulberry trees and provide references in its industrial use. This paper established a simple, precise and accurate method to rapid determine the content of DNJ in mulberry resources and other related products. This paper also investigates technology to purify all the PHAs in silkworm excrement, mulberry leaves and mulberry twigs.1. Study on determination of the content of DNJ, in mulberry resources by HPLC-ELSDThis section mainly focused on determining the content of DNJ in mulberry leaves, branch bark, branch xylem, twigs(one year old), stems (one year old), stem xylem (more than two years old), root barks, root xylem in three trees of Morus australis and mulberry leaves, branch bark, branch xylem, root barks, root xylem in three trees of Morus alba L. by high-performance liquid chromatography coupled to an evaporative light scattering detector (ELSD). The method was established using TSKgel Amide-80 (4.6 mm×250 mm,5μm) column with acetonitrile-water (84:16) as mobile phase and the water containing 6.5 mM ammonium acetate. The flow rate was 1.00 mL/min, column temperature 30℃. The temperature of the drift tube was set at 55℃, gas flow rate:3.2 L/min. the gain was usually set at 8. injection volume: 10μL. Standard curve for DNJ using ELSD was linear in the range of 0.505μg~20.2μg, r=0.9991, n=6. The average recovery was 94.95%, n=5, RSD=1.98. The method is accurate, reproducible. It can be used for the rapid determination of 1-deoxynojirimycin in mulberry resources and other related products. 2. Study on technology of isolating DNJ and Fagomine, and Purifing PHAs in silkworm shit, mulberry leaves and mulberry twigs.Using strongly acidic cation exchange resins (732), strongly basic anion exchange resin (201×7), weakly acidic cation exchange resin (HD-2), macroporous resin, Sephadex LH-20, alcohol precipitation to isolate DNJ and Fagomine, and Purify PHAs in silkworm shit, mulberry leaves and mulberry twigs.
