The effect of bone marrow mobilization combined with Vitamin D3 on diabetic mice fundus.Purpose:Investigating the effect of Vitamin D3 (VD3), bone marrow mobilization and bone marrow mobilization combined with VD3 on experimental diabetic mice fundus.Method:32 mice (C57/BL6J rat weighting for 20-25g), After glucose analysis,3 healthy mice were chosed for blank control(E group). The others were diabetic with 2%STZ (streptozotocin) (175 mg/kg intraperitoneally in citrate buffer) after abstinence from food for 15h. Blood was obtained from a tail vein to make glucose analysis 3 times in the third day after STZ Injection (blood glucose concentration> 16.7 mmol/L as diabetic model). Randomly distributed into four groups:VD3 group (A group), bone marrow mobilization group(B group), combination therapy group(C group), control group (D group). In 14th day after STZ injection, A and B group animals were treated with 1,25-(OH)2D35 mg/kg intraperitoneally once a week. In 28th day after STZ injection, B and C group animals were treated with rhG-CSF 50μg/kg subcutaneous injection once daily, consecutive five days. At the 7th,14th,21st,28th,35th,42nd,49th day, we take blood sugar and Ca2+ monitoring, direct ophthalmoscope examination, thickness measurement by pathologic analyzing software. Processing data by SPSS 19.0.Result:Choroid membrane of D group atrophy lasted from 14th day to 21th day (F=437.411, P=0.000). Edema of the retina began from 7th day (P=0.000), but consistently atrophy over time (F=22.338,P=0.003). VD3 had protective effect to choroid membrane (P=0.003), but didn't have effect to atrophy of the retina. B group and C group both have protective effect to choroid membrane and retina (P<0.05). HE staining indicated that retinal neovascularization in diabetic mice had form from 14th day. Fluorescein angiographyh (FFA) indicated that microaneurysm appeared from 14th day and hemorrhage became obviously from 28th day in all group. Hemorrhage in A and B group lasted to 49th day, but C group hadn't have hemorrhage since 35th day. The hemorrhage in C group was the least in all groups(x2=9.22, P=0.026). FFA indicated that retinal neovascularization appeared from 42th day in diabetic mice in D group, but didn't appear in any other group.Conclusion:Both Vitamin D3 and bone marrow mobilization combined with Vitamin D3 have protective effect on diabetic mice choroid membrane. Combined treatment had protective effect on atrophy of retina of diabetic mice, but Vitamin D3 didn't have the effect. Bone marrow mobilization had protective effect on atrophy of choroid membrane and retina. Both Vitamin D3 and bone marrow mobilization could effectively inhibited hyperpermeability of the retinal vessels separately.
