Objective:Taking HL60 cells as researching objects, discuss the effect of dihydroartemisinin(DHA) combination of daunorubicin(DNR) on HL60 Cells.Methods:HL60 cells are cultured in vitro. By using techniques of the colorimetric 5-diphenyl bromide (MTT), the effects of DHA, DNR and DHA combination of DNR on proliferation of HL60 cells are detected. After effects of DHA and DHA combination of DNR on HL60 cells, using flow cytometry to detect the cells'rate of apoptosis and oncosis, and using transmission electron microscope to observe the changes of cell morphology.Result:DHA in dose of 12.5-50μg/ml had obvious inhibition on the proliferation of HL60 cells in this experimental research, which were dose-time dependent. DNR in dose of 2.5-10μg/ml had obvious inhibition on the proliferation of HL60 cells, but which were not dose-time dependent. 50μg/ml DHA combination of 10μg/ml DNR had better inhibition on the proliferation of HL60 cells than the single drug groups. DHA kill HL60 cells by inducing apoptosis and oncosis. DHA in dose of 12.5μg/ml, the main way of cell death is apoptosis, and DHA in dose of 25 or 50μg/ml, is oncosis. The oncosis rate of HL60 cell is proportional to the dose of DHA.50μg/ml DHA combination of 10μg/ml DNR kill HL60 cells by inducing oncosis.Under Transmission electron microscopy,it could be seen that:HL60 of DHA high dose group had tow ways of apoptosis and oncosis to change, and necrosis was the main change. HL60 of DHA high dose combination of DNR high dose group had only one way of oncosis to change.Conclusion:DHA could inhibit proliferation of HL60 cells by inducing apoptosis and oncosis., which was dose-time dependent in a certain range of concentrations. The oncosis rate of HL60 cell is proportional to the dose of DHA. DHA combination of DNR inhibited proliferation of synergy or antagonism to HL60 cells.50μg/ml DHA combination of 10μg/ml DNR could inhibit proliferation of HL60 cells by inducing oncosis, which was synergy.
